  • Study on Part of Key Techniques of Standardized Cultivation about Chinese Traditional Medicine Platycodon grandiflorum (Jacq.) A.

    [Objective] Based on GAP authentication, the standardized cultivation of Chinese traditional medicine Platycodon grandiflorum (Jacq.) A. in North China was studied in order to ensure the medicinal plants from the source. [Method] The best harvest time of Platycodon grandiflorum (Jacq.) A. was confirmed, and the pollution-free precautionary method of controlling Seedling Blight and Leaf Spot was obtained. [Result] The results showed that the best harvest time of Platycodon grandiflorum (Jacq.) A. was in the second ten days of September when the cool and dry climate was convenient for the post-processing. It had good effects of applying 75% 0.7 g/L Jubang ultramicro wet powder and 50% 1.4 g/L Carbendazim WP ultrafine powder simultaneously to control Seedling Blight and Leaf Spot of Platycodon grandiflorum (Jacq.) A.. [Conclusion] The study has laid the theoretical basis for high-yield cultivation of Platycodon grandiflorum (Jacq.) A. .

  • Effects of IBA on the Hydroponics of Nanocnide lobata

    [Objective]To provide technical support for breeding of Nanocnide lobata by screening the optimum concentration of IBA.[Method]With rooting rate,adventitious root amount,root length,lateral root number and developing index of root as indicators,the root situation of Nanocnide lobata cuttings treated with IBA at the concentration of 0,30,60 and 90 mg/L and treating time of 2,4 and 6 h were tested respectively.[Result]The results showed that the appropriate concentration and soaking time of IBA were 60 mg/L and 6 h respectively,under which the lateral root number,adventitious root number,adventitious root length and the developing index of root were 16.6,48.6,15.8 mm and 424.9 respectively.[Conclusion]The study is of great importance in instructing the breeding of Nanocnide lobata.

  • Effects of Soaking with Paclobutrazol on Seed Germination and Seedling Growth of Cassia obtusifolia L.

    [Objective] The research aimed to study the effects of soaking with paclobutrazol on seeds germination and seelding Growth of Cassia obtusifolia L.[Method] The seeds of Cassia obtusifolia L.were soaked by different concentration solution of paclobutrazol in different hours.[Result] The results showed that soaking Cassia obtusifolia L.seeds with pacbutraol delayed the germination of seeds,reduced the seed germination percentage and the germination energy,but it could increase the concise seed vigor index.And soaking Cassia obtusifolia L.seeds with paclobutraol increased the stem diameter,dwarfed the plant,induced lateral roots growth and enhanced obviously the lodging resistance of seeding.Among those of the control,the effect of 100 mg/L paclobutraol soaking Cassia obtusifolia L.seeds in 24 h was the best.[Conclusion] The study can provide theoretical basis for high yield cultivation of Cassia obtusifolia L.

  • Investigation and Floristic Study on the Medicinal Seed Plants in East Guangdong Province

    The species diversity of medicinal seed plants in East Guangdong Province were investigated,and the compositions and distribution types of the families and genera were analyzed too.The results showed that:①There were 1 132 species of medicinal seed plants in the area belonging to 614 genera of 159 families,of which 10 species of the medicinal plants belonging to 9 genera of 6 families were gymnosperms,and 1 142 species belonging to 605 genera of 153 families were angiosperms;②The dominant families were distinct and the diversity of species were rich;③There were 15 distribution types of the medicinal seed plants in this area,of which the tropical floristic elements were absolute predominance,and there was a few endemic elements in the area;④In the area,the medicinal resources were various,and some floras were ancient and fragile.

  • Textual Research and Surveying of Officinal Varieties on Arisaematis Rhizoma

    [Objective] Through textual research and surveying of officinal varieties in Arisaematis Rhizoma,we expected to offer some reference for its rational exploitation.[Methods] We unscrambled officinal varieties of Arisaematis Rhizoma in ancient books and records about chinese medicine,and combined some classification records to researched its synonym,error citation as well.We made some market survey about its supply state.[Results] We considered Arisaematis Rhizoma name is in ceaseless change and its original is in confusion in the development.The original plants of Arisaematis Rhizoma are from many species and have wide distribution.There are 38 species original plant as Arisaematis Rhizoma in nationwide.they largely concentrate upon Arisaema.There are also Pinellia,Typhonium,Amorphophallus except Arisaema.Through on-site inspection in Anhui,Hebei,Gansu,Gangdong and so on,we found Arisaematis Rhizoma mainly is Pinellia pedatisecta in market,the species in pharmacopeia all are from wild and have a little rate in market.All the seedlings of Arisaematis Rhizoma are Pinellia pedatisecta in market by author identification.[Conclusion] Arisaematis Rhizoma is poisonous and contains rank poison components in some species.The resemble appearance in various species lead its complicated and disordered original state,which not only affect its curative effect,but also influence clinical security.Government departments should put an important attachment to it.

  • Study on Extraction and Purification Technology of Chlorogenic acid from Lonicera japonica Thunb.

    [Objective] The aim was to study the extraction and purification technology of Lonicera japonica Thunb..[Method]The optimum extraction and purification technologies were obtained from the single factor test and the orthogonal test respectively.[Result]The optimum extraction conditions were as follows:extraction temperature of 60 ℃,pH of 4,ethanol concentration of 60%,material-liquid ratio of 1:25 and extraction time of 1.5 h,under the above conditions the yield of chlorogenic acid was 4.6%.The optimum purification conditions were as follows:eluent concentration of 20%,eluent volume of 5 BV,eluent pH of 8 and flow rate of 5 ml/min,with which the purity of chlorogenic acid was 58.3%.[Conclusion]The optimum extraction and purification technology were obtained,which would provide experimental evidence for the industrialized production of chlorogenic acid.

  • Study on Habitat Processing of Radix platycodonis

    [Objective]The study aimed to confirm the effects of peeling and drying temperature on polysaccharides content of Radix platycodonis,and providing reference for establishing the best habitat processing of it.[Method]Extracted with water,refined with ethanol,the contents of polysaccharides in Radix platycodonis were determined by phenol-H2SO4 colorimetry method.[Result]The polysaccharides content between the unpeeled sample and the peel varied a little,both of which were lower than that in the peeled sample.The drying speed of the peeled samples was promoted,and the content of polysaccharides was the highest dried at 50 ℃.[Conclusion]As the content of polysaccharides in peeled sample increased 6.1%,and peeled sample was easier to dry,peeling was screened as the final processing method.

  • Study on the Population Diversity of Endophytes in Tibetan Endemic Sophora moorcroftiana

    [Objective]To discuss the population diversity of endophytes in Sophora moorcroftiana from Tibet.[Method]Separated out from the root,stem,leaf and seed of Sophora moorcroftiana,the endophytes were observed with optical microscope.[Result]Sixty-eight endophytes were obtained by morphological observation,which belonged to 14 genera,7 families,6 orders and 4 classes.The amount,distribution,population formation of endophytes from different organs of Sophora moorcroftiana and from Sophora moorcroftiana in different habitats varied.[Conclusion]The study provided theoretical evidence for developing and utilizing Sophora moorcroftiana resources.

  • Study on Dryopteridaceae of Medicinal Pteridophyte in Huangshan Fuxi Valley

    [Objective] The aim was to study species,distribution,medicinal value and utilization of medicinal pteridophyte of Dryopteridaceae in Huangshan FuxiValley.[Method] Through primary investigation on pteridophyte resources in Huangshan Fuxi valley,combined with the utilization situation in southwest minorities,the main species and distribution of Dryopteridaceae in medicinal Pteridophyte resources in Fuxi valley were evaluated.[Result] The results showed that there are 12 species of Pteridophyte,which belong to 3 genera.The protection and utilization prospects were also discussed.[Conclusion] The study will provide data for further development and safety clinical application of Pteridophyte resources.

  • Analysis of Ether-soluble Components in Ginseng,Red Ginseng and Steamed Ginseng Water by GC-MS

    [Objective]The study aimed at providing reference for the development and utilization of ginseng products and the by-products through analyzing the ether soluble components in fresh ginseng,red ginseng and steamed ginseng water.[Method]GC-MS was used to analyze the components.[Result]The 94,95 and 24 compounds were separated from fresh ginseng,red ginseng and steamed ginseng water respectively,of which the 31,33 and 10 compounds were identified respectively.Meanwhile,there were 15 compounds identical in fresh ginseng and red ginseng.The falcarinol of anti-cancer activity and squalene of immune competence were firstly identified in fresh ginseng and red ginseng.[Conclusion]The ether soluble components in fresh ginseng,red ginseng and steamed ginseng water are comparatively analyzed in the study,which will provide reference for development and utilization of ginseng products and the by-products.

  • Determination of Total Flavonoids in Limonium aureum(L.) Hill.by Spectrophotometry

    [Objective] The aim was to establish a simple and quick method to determine total flavonoids content in Limonium aureum(L.) Hill.and provide scientific evidence for the quality control and development of Limonium aureum(L.) Hill.[Methods] Quercetin as reference substance,UV spectrophotometry was adopted in this study and the detection wavelength was 370 nm.[Results] The linear relationship was good(r=0.999 6) in the range of 2.5-12.5 μg/ml,the average recovery rate of total flavonoids was 99.46% and the content of total flavonoids in the samples was 1.63%.[Conclusion] UV spectrophotometry is a suitable method for detecting total flavonoids content in Limonium aureum(L.) Hill.with the advantages of simple and quick which would provide the scientific evidence for the quality control and development of Limonium aureum(L.) Hill.

  • Isolation of Endophytes from Sophora alopecuroides and Screening of Antagonistic Strains

    [Objective]The aim was to screen the antagonistic strains isolated from wild Sophora alopecuroides in Ningxia.[Method] The streak plate method was used to isolate the endophytes,and the duel-growth method was used to screen the antagonistic strains.[Result]Kinds and amount of endophytes from Sophora alopecuroides in different origins and different parts of the same plant varied.One hundred and eighty entophytic strains were obtained from Sophora alopecuroides,of which ten antagonistic strains were obtained.[Conclusion]The entophytic strains and antagonistic strains are obtained respectively in this research,which is of important value in utilizing the Sophora alopecuroides rationally.

  • Analysis on Chemical Components of the Volatile Oil from Senecio jacobacea L.by GC-MS

    [Objective]The research aimed at analyzing the chemical components and their relative content of volatile oil from Senecio jacobacea L.[Method]The volatile oil was extracted from Senecio jacobacea L.with steam distillation and analyzed by GC-MS.[Result]Thirty compounds were identified from the volatile oil,and the major compositions were aromadendrene oxide-(2)(22.17%),aristolene epoxide(11.25%),caryophyllene oxide(7.56%),7R,8R-8-hydroxy-4-isopropylidene-7-methylbicyclo[5.3.1]undec-1-ene(7.36%),(-)-spathulenol(6.01%),and aromadendrene oxide-(1)(5.35%).[Conclusion]The chemical components and their relative content of volatile oil from Senecio jacobacea L.were obtained,which would provide the basis for comprehensive development and utilization of Senecio jacobacea L..

  • Study on Extraction of Polyphenols from Punica granatum Linn.Peel and Its Antibacterial Activity

    [Objective]To optimize the extraction process of polyphenols from Punica granatum Linn.peel and discuss the antibacterial activity of the crude extracts.[Method]Microwave-assisted extraction(MAE) was used to extract polyphenols,and the extraction process was optimized by employing both the single-factor test and orthogonal test.The inhibitory activity of the crude extracts on Staphylococcus aureus,Proteus vulgaris and Bacillus cereus were compared.[Result]The optimum extraction conditions were as follows:screen of 60-80 mesh,extraction power of 385 W,solid-liquid ratio of 1:25(W/V,the same as following),extraction duration of 120 s and extraction times of 3,under which the yield of polyphenol from Punica granatum Linn.peel was 21.4%.The inhibitory activity of 0.96 mg/ml extracts on Staphylococcus aureus,Proteus vulgaris and Bacillus cereus were the strongest,the corresponding diameter of inhibition zone were 1.15,1.10 and 0.95 cm respectively and the corresponding MIC were 0.80,1.60 and 0.40 g/ml respectively.[Conclusion]The natural medicine was extracted from Punica granatum Linn.peel,which provided theoretical evidence for exploiting natural bacteriostat.

  • Study on Chemical Composition of Volatile Extracts from Different Parts of Jatropha curcas L.by Supercritical Fluid Carbon Dioxide Extraction Technique

    [Objective]The aim was to investigate the kinds of chemical components and their content in leaf,bark and seed extracts of Jatropha curcas L.by supercritical CO2 extraction,aiming at providing basis for the development of Jatropha curcas L.in the field of pesticide and medicine.[Method] Supercritical fluid CO2 extraction technique was used to obtain volatile components from different parts of Jatropha curcas L..The chemical constituents of extracts were determined by GC-MS.[Result]The extracts were deep yellow or light yellow,and the yield of leaf,bark and seed were 0.53%,0.56% and 0.78% respectively.The 26,26 and 17 chemical components were obtained from leaves,bark and seeds of Jatropha curcas L.respectively,which took up 66.88%,36.44% and 88.31% of total volatile components respectively.The main components were ketones,esters,fats,terpenoids,steroid alcohols,organic acids and alkanes etc.The results showed that there was an evident difference in chemical composition and relative content of extracts from different parts of Jatropha curcas L..[Conclusion] The study will provide basis for further understanding of chemical composition in different parts of Jatropha curcas L.and provide reference for its development in the field of medicine and pesticide etc.

  • Comparative Research on the Essential Oil from Senecio scandens

    [Objective]The aim was to study the chemical components and relative content of the essential oil from the cultivated and the wild Senecio scandens,expecting to provide the scientific evidence for the reasonable utilization of Senecio scandens.[Method]The essential oil was extracted from the cultivated and the wild Senecio scandens with the steam distillation,and was analyzed with GC-MS.[Result]The results showed that main chemical components of the cultivated-essential oil and the wild-essential oil had certain difference:the 23 compounds and 21 compounds were identified from the cultivated and the wild Senecio scandens respectively,the 13 compounds were the same among them.The cultivated-essential oil contains 17 terpenoids,6 aliphatic and aromatic compounds,while the wild-essential oil contains 9 terpenoids,12 aliphatic and aromatic compounds.The relative content was different:the main components of essential oil from the cultivated Senecio scandens were 11.60% of α-farnesene,13.49% of α-caryophyllene,8.57% of caryophyllene oxide,10.86% of palmitic acid and 9.00% of linoleic acid,while that of essential oil from the wild Senecio scandens were 8.10% of α-farnesene,19.50% of α-caryophyllene,14.22% of caryophyllene oxide,21.45% of palmitic acid and 13.66% of linoleic acid.[Conclusion]The chemical components of essential oil in Senecio scandens was identified and each content was determined by this method,thus the study will provide the scientific evidence for the reasonable exploitation and sustainable utilization of Senecio scandens.

  • Effects of Different Growth-regulators on Shoot Regeneration Capacity of Dioscorea oppasita

    [Objective]To provide basic data for establishing a virus-free system for stem tips of Dioscorea oppasita by studying the comprehensive effects of different growth-regulators with different concentration on the shoot regeneration capacity.[Method]With 6-BA,IBA,KT and CH as independent variables and differentiation rate,proliferation coefficient(total amount of regenerative shoots/total amount of explants) and proliferation rate as dependent variables,nine formulas were designed with MS as basic medium to study the regeneration capacity of Dioscorea oppasita shoots.[Result]The contribution efficiency of 4 growth-regulators on the proliferation coefficient and proliferation rate were 88.1% and 92.4% respectively and the effect size was 6-BA>IBA>CH>KT.[Conclusion]The effects of 6-BA and IBA on the regeneration capacity(proliferation coefficient or proliferation rate) of Dioscorea oppasita shoots were strong.

  • SSR Analysis of Genetic Diversity of Panax ginseng

    [Objective] The differences among the ginseng germplasm resources in five different producing regions of Jilin Province were analyzed,so as to provide theoretical basis for the introduction of ginseng from other places.[Method] By using simple sequence repeat(SSR),nine ginseng materials of five districts in Jilin were used for genomic polymorphic analysis.The dissimilarity of coefficient was calculated by professional software DPS 9.50.And the dendrogram was made by using Nei-Li formula and UPGMA.[Result] 16 pairs of polymorphic primers were selected from 25 pairs of primers for the amplification.181 bands were amplified and 117 bands were polymorphic,with the polymorphic percentage of 64.7%.[Conclusion] There was much genetic diversity at the molecular level among the germplasm resources of Panax ginseng actually.The differences between the tested ginseng variteal types were determined by the genetic factors.Furthermore,it was also affected by environmental factors.

  • Study on the Reducing Blood Lipid and Antioxidation Effects of Flavonoids from Eucommia ulmidoes Olirer Leaves

    [Objective] The paper aimed to further observe the activity of reducing blood lipid and antioxidation effects of flavonoids from Eucommia ulmidoes Olirer leaves.[Method] ICR mice were randomly divided into the normal control group and high-fat diet group,the normal control group was fed by common feedstuffs,while high-fat diet group fed by high-fat diet,which made into tested mice of the nutrition,hyperlipidemia model.And high-fat diet groups were randomly divided into model control group,positive medicine control group,flavonoids large,medium and small-dose groups were taken medicine by flavonoids gavage with 45 mg/kg·d,15 mg/kg·d and 5 mg/kg·d dose,positive drug group was offered by Lovastatin with 5 mg/kg·d(equivalent to human dose conversion according to the standard body surface area of mice with 2 times doses),the normal control group was offered by equal volume of solvent.After 17 d,each group of mice was fasted by 12 h,orbital blood,heparin,0.2 ml of blood from each of hemolysis for the CAT after the determination of the rest of the blood and anticoagulant used for centrifugal separation of plasma lipids and POD,MDA determination.[Result] Compared with hyperlipidemia model group,TC,TG,LDL-C and AI flavonoids from Flavonoids from E.ulmidoes Olirer leaves had degraded with different degrees,HDL-C had raised,activity of CAT,POD had strengthened,content of MDA in plasma had degraded.[Conclusion] Flavonoids from E.ulmidoes Olirer leaves had effects of reducing the nutritional hyperlipemia in mice and anti-lipid oxidation.

  • Isolation of Genomic DNA in Morinda citrifolia Linn.and Establishment of Its ISSR-PCR Reaction System

    [Objective] The research aimed to isolate the genomic DNA in Morinda citrifolia Linn.and establish its ISSR-PCR reaction system.[Method] The Morinda citrifolia Linn.leaves of 3 germplasm resources from Hainan and 4 germplasm resources from America were chosen as test materials,the isolation of DNA in Morinda citrifolia Linn.and establishment of ISSR-PCR were studied.[Result] The results showed that DNA of Morinda citrifolia Linn.leaves with high quality were gained by the modified CTAB method.The isolated DNA could be used for ISSR analysis and 7 primers were found to amplify polymorphic products.[Conclusion]The rapid and efficient extraction method and the ISSR analysis procedure in this experiment have laid a good foundation for application of ISSR in Morinda citrifolia Linn.genetic research.

  • RAPD Analysis of Seven Medical Asteraceae Plants

    [Objective] The research aimed to analyze 7 medical Asteraceae plants by random amplified polymorphic DNA(RAPD).[Method] 10 stable polymorphic primers were screened from 50 primers which were used for amplification,the phylogenetic tree was established by UPGMA clustering procedure on the basis of amplified DNA fragment.[Result] 337 clear polymorphic bands were obtained.Clustering analysis showed that the phylogenetic tree established by RAPD molecular marker was identified with traditional classification.[Conclusion] The study can provide reference for identifying medical plants of Asteraceae and laying basis for the pertinence protection of that.

  • Determination of Ursolic Acid and Oleanolic Acid Contents in Kuding Tea of Aquifoliaceae

    [Objective] The research aimed at determining the contents of ursolic acid and oleanolic acid in different species and different origins of Kuding tea and providing basic data for the industrialization development of ursolic acid.[Method] The contents of ursolic acid and oleanolic acid in functional leaves from 6 species of Kuding tea in Aquifoliaceae and 4 kinds of the commercial Ilex kudingcha C.J.Tseng shoots were determined by high performance liquid chromatography(HPLC).The chromatographic conditions were as follows:Eclipse XDB-C18 column(5 μm,4.6 mm×150 mm),mobile phases of methanol-0.2% aqueous solution of phosphoric acid(v:v=88:12),detection wavelength of 220 nm,flow rate of 0.8 ml/min,column temperature of 35 ℃ and injection volume of 20 μl.[Result] The contents of ursolic acid from 6 species of Kuding tea in Aquifoliaceae were as follows:I.cornuta Lindl.(1.64%)>I.huoshanesis Y.H.He(1.63%)>I.latifolia Thunb.(1.54%)>I.pentagona S.K.Chen,Y.X.Feng et C.F.Liang(1.23%)>I.kudingcha C.J.Tseng(0.96%)>I.centrochinesis X.Y.Hu(0.69%)>the shoots of Ilex kudingcha C.J.Tseng(<0.28%).The contents of oleanolic acid from 6 species of Kuding tea in Aquifoliaceae were as follows:I.latifolia Thunb.(0.22%)>I.huoshanesis Y.H.He(0.18%)>I.cornuta Lindl.(0.17%)>I.pentagona S.K.Chen,Y.X.Feng et C.F.Liang(0.14%)>I.kudingcha C.J.Tseng(<0.12%)>I.centrochinesis X.Y.Hu(0.1%)>the shoots of Ilex kudingcha C.J.Tseng(0).[Conclusion] The determination results were favorable for the selection of suitable materials for the processing of ursolic acid.

  • Study on Fingerprints of Alpinia oxyphylla Miq.by HPLC

    [Objective]The aim was to establish the fingerprints of Alpinia oxyphylla Miq.by HPLC.[Method]The HPLC was used and the chromatographic conditions were as follows:column of ODS Hypersil VP-ODS(150 L×4.6 mm),mobile phases of water-acetonitrile(V:V=70:30),sampling volume of 20 μl and detection wavelength of 220 nm.[Result]The fingerprints of Alpinia oxyphylla Miq.obtained from different areas of Hainan Province were similar,while different from that obtained from Guangdong Province and Guangxi Province.The fingerprints of Alpinia oxyphylla Miq.obtained from Guangdong Province and Guangxi Province were similar as that obtained from the other areas.[Conclusion]Ten kinds of fingerprints were obtained by this rapid,convenient and reliable method,which would be a new rapid identification method for Alpinia oxyphylla Miq..

  • Simultaneous Determination of 9 Nucleosides analogues in Fritillaria anhuiensis by RP-HPLC

    [Objective] To develop reverse phase high performance liquid chromatography(RP-HPLC) method for simultaneous determination of 9 nucleosides and nucleobases in Fritillaria anhuiensis S.C.Chen et S.E.Yin.[Methods] The analyses were performed on an Agilent Zorbax SB-Aq C18(4.6 mm × 250 mm,5 μm)for all analyses.The investigated compounds were separated with gradient mobile phase consisting of water and Methanol.The flow rate was 1.0 ml/min.The detection wavelength was 260 nm with the 25 ℃ temperature of column.[Result] The investigated compounds including uracil,cytidine,uridine,hypoxanthine,inosine,guanosine,thymidine,adenosine,adenine had good linearity(r2 ≥ 0.998 2).The average recovery was 96.7% to 104.3% with RSD ≤ 3.87%.[Conclusion] The accuracy,stability,repeatability,and average recovery of the method are satisfying,and the 9 nucleosides and nucleobases of F.anhuiensis can be rapidly and accurately quantified by RP-HPLC.

  • Analysis of Fatty acid Components in Different Strains Panax ginseng C.A.Mey.by GC-MS

    [Objective]The aim was to provide an important index on breeding of Panax ginseng C.A.Mey.by determining the constituents and relative content of the fatty acid from different strains of it.[Method]The soxhlet extraction and the GC-MS were used.[Result]The results showed that 20 compounds in the fatty acid from the Ginseng were isolated and identified.The major components are(Z,Z)-9,12-Octadecadienoic acid,hexadecanoic acid and(Z,Z,Z)-9,12,15-Octadecatrienoic acid,but the content was different:①The(Z,Z)-9,12-Octadecadienoic acid is of 47.896% in damaya,50.901% in ermaya,50.254% in yuanbangyuanlu and 49.456% in changbo,the content is highest in ermaya.②The hexadecanoic acid is of 12.370% in damaya,12.535% in ermaya,12.614% in yuanbangyuanlu and 11.983% in changbo,the content is highest in yuanbangyuanlu.③The(Z,Z,Z)-9,12,15-Octadecatrienoic acid is of 10.407% in damaya,12.487% in ermaya,12.026% in yuanbangyuanlu and 12.026% in changbo,the content is highest in ermaya.[Conclusion]The method can identify the different strainsof Panax ginseng C.A.Mey and it may be an important tool on breeding of ginseng.

  • Effect of ZTC1+1-Ⅱ Clarifying Agent on Flocculation Process of Alcohol-extract Solution from Ginkgo biloba Linn.Leaves

    [Objective] The ZTC1+1-Ⅱ natural clarifying agent was used to flocculate alcohol-extract solution of Ginkgo biloba Linn.leaves for optimizing the production process and improving the clarity and quality of solution.[Method] There are two kinds of clarifying agent,named A and B,which are formulated by distilled water or 1% acetum and pre-swelled for 24 h then filtrated with double layer gauze.The weight of flocculation was used as the main parameters to estimate the effects of different factors on flocculation.[Result] The best flocculation technique conditions were as follows:the adding sequence of clarifying agent was clarifier B before clarifier A,the pH value of alcohol-extract solution was 5.5,the optimum amount of clarifier B and A was 1.6 and 0.8 g/L,working temperature of clarifying agent was 40 ℃,clarifying time was 40 min.[Conclusion] The study can reduce the follow-up processing and the production cost.

  • Study on the Role in Scavenging Reactive Oxygen Free Radicals of Citrus Flavonoides in Vitro

    [Object] The research aimed to study the role in scavenging reactive oxygen free radicals of Citrus flavonoids in vitro.[Method] The rate of Pyrogallol autoxidation was determined after adding Citrus flavonoids,the role in scavenging reactive oxygen free radicals of Citrus flavonoids in vitro was studied,and it was compared with the roles of vitamin C and 75% alcohol.[Result] The average inhibition rates of 0.1 ml 1 mg/ml vitamin C and 0.1 ml 1 mg/ml Citrus flavonoids to 45 mmol/L Pyrogallol autoxidation reached 50.98%±13.9%,27.8%±2.58%,there existed extreme difference(P<0.01).The average inhibition rate of 0.1 ml 75% alcohol to 45 mmol/L Pyrogallol autoxidation reached 10.89%±1.84%,and it was compared with 0.1 ml 1 mg/ml Citrus flavonoids and 0.1 ml 1 mg/ml vitamin C,there existed extreme difference(P<0.01).[Conclusion] Citrus flavonoids played an obviously role in scavenging reactive oxygen free radicals of Citrus flavonoids in vitro.

  • Optimization of Extraction Process of Compound Traditional Chinese Medicine by Orthogonal Design

    [Objective] The objective was to screen the best extraction process of compound traditional Chinese medicine that is composed of Radix Sophorae Flavescentis,Rhizoma Smilacis glabrae and Cortex Dictamni and provide experimental evidence for the development and utilization of the compound prescription.[Method] The minimal inhibitory concentration(MIC) of extracts on Candida albicans as index,the orthogonal test was designed with 4 factors:concentration of ethanol(A),extraction time(B),the proportion of three components(C) and material-liquid ratio(D) to screen the optimum extraction conditions.[Result] The optimum technology was as follows:extraction solution of 70% ethanol(V/V),proportion of Radix Sophorae Flavescentis,Rhizoma Smilacis glabrae and Cortex Dictamni was 2:2:1,material-liquid ratio of 1:10 and extraction time of 2 h,under the above conditions the yield of the compound traditional Chinese medicine was 23% and the MIC of Candida albicans was 0.195 mg/ml.[Conclusion] The optimum extraction technology was obtained from the orthogonal test,which would provide reference for the development and utilization of the compound traditional Chinese medicine.

  • Preparation for Healthy Tablet of Plantago asiatica L.

    [Objective] The research aimed to study the preparation process for healthy tablet of Plantago asiatica L..[Method] Using concentrated solution of Plantago asiatica L.,maltodextrin,mannitol,aspartame and sorbitol as main materials,healthy tablet of Plantago asiatica L.was prepared by the process of mixing,prilling,drying and tabletting and so on.[Result] The best extracting process of Plantago asiatica L.extract was as follows:85% ethanol as extractant and extracted by ultrasonic for 30,20 min at 65 ℃,the ratio of material to liquid was 1:6 and 1:4 respectively.0.2% β-cyclodextrin could improve the taste of Plantago asiatica L.extract.The formula of Plantago asiatica L.tablet was as follows:50 ml 50% Plantago asiatica L.extract 70 g filter,3 ml 5% menthol-ethanol,35 ml 10% polyvinylpyrrolidone(PVP).[Conclusion] The prepared Plantago asiatica L.tablet is of good tasty,delicate and moderate sweet,low sugar,low calorie and unique flavor and so on.

  • Preliminary Study on HPLC Fingerprint of Artichoke(Cynara scolymus L.) Leaves

    [Objective] To establish the HPLC fingerprint of Cynara scolymus L.leaves extracts.[Methods] The Cynara scolymus L.leaves were extracted with 70% ethanol(V/V) and analyzed by HPLC,the chromatographic condition were as follows:C18 column(4.6 mm×250 mm,5 μm),mobile phases of acetonitrile-0.5% acetic acid-water,gradient elution,detection wavelength of 330 nm.[Results] 14 batches of the extracts were analyzed by Chinese Medicine Chromatographic Fingerprint Similarity Evaluation software,and 15 peaks were identified as fingerprint.The common mode of Cynara scolymus L.leaves extracts was preliminary established.[Conclusion] The fingerprint established by this method will provide basic data for the quality control of artichoke leaves.

  • Distribution and Accumulation Characteristics of Heavy Metals in Different Organs of Mentha haplocalyx in Guizhou

    [Objective] The aim is to study distribution and accumulation characteristics of five heavy metals(Pb,Cd,Hg,As and Cu) in different organs of Mentha haplocalyx and discuss factors that affecting heavy metals in the herb,which will provide some valuable information for selection certain organs of Mentha haplocalyx for medical uses.[Method] Atomic absorption spectrometer and atomic fluorescence spectrometer were adopted to determine the concentrations of heavy metals.[Results] Concentrations of As,Hg and Pb in the plant organs showed the following order:leaf>root>stem,the order of concentration for Cu was:root>leaf>stem,while the order of the concentration for Cd was:root>stem>leaf.The concentrations of heavy metals displayed some obvious differences in Mentha haplocalyx leaves(r=-0.952,P<0.05).The concentrations of Pb and Cu in soils of GAP base showed a negative correlation(r=-0.954,P<0.05).As and Cu in these GAP soils also exhibited a strong negative correlation(r=-0.990,P<0.05),while As and Pb in the soil demonstrated a strong positive correlation(r=0.987,P<0.05).[Conclusion] The concentrations of heavy metals displayed some abvious differences in different organs of Mentha haplocalyx while there were no significant correlations between the concentrations of heavy metals in the GAP soil and those in the organs of Mentha haplocalyx.

  • Optimization of Extraction Process of Polysaccharides from Fruiting body of Cordyceps militaris L.Link by Orthogonal Test

    [Objective]The research aimed to optimize the extraction process of polysaccharides from fruiting body of cultured Cordyceps militaris L.Link by orthogonal test.[Method] Decocting extraction method,hydrothermal refluxing method and alkali extraction method were used to extract polysaccharides from fruiting body of cultured Cordyceps militaris L.Link respectively.[Result]The optimum conditions of decocting extraction method were as follows:solid-liquid ratio of 1:40,extraction time of 3.0 h and extraction times of 3,and the effects order of 3 factors on the yield of polysaccharides was solid-liquid ratio> decocting time> decocting times.The optimum conditions of hydrothermal refluxing method were as follows:the sample powder was mixed with water at ratio of 1/20 and refluxed for 1.0 h,2 duplicates,and the effects order of 4 factors on the yield of polysaccharides was extraction times> extraction time> extraction temperature> solid-liquid ratio.The optimum conditions of alkali extraction method were as follows:extracting solution of 0.7 mol/L NaOH,solid-liquid ratio of 1:8,extraction duration of 0.5 h and extraction times of 3,and the effects order of 4 factors on the yield of polysaccharides was extraction times> concentration of NaOH> solid-liquid ratio> extraction time.[Conclusion]The optimum extraction process of decocting extraction method,hydrothermal refluxing extracting method and alkali extracting method were obtained in this study,which provided reference for the further research and industrial production of fruiting body from cultured Cordyceps militaris L.Link.
