  • A Comparison of Leaf Morphological Characters among Different Major Cultivars of Rehmannia glutinosa Li-bosch

    [Objective] To qualitatively identify 8 major cultivars of Rehmannia glutinosa Libosch by comparing their leaf characters, including "85-5", "Beijing No.1", "Beijing No.3", "Shengjin", "Qinhuai", "Huaidi No.1", "Huaidi No.2" and "Huaidi No.3", so as to provide theoretical evidences for the identification and selective breeding of major cultivars. [Method] The length and width of R. glutinosa leaves were measured by using a ruler. And the density and length of the non-glandular hair, the density, size and density of the stomata in the epidermis were measured under a scanning electron microscope. [Result] The results showed that there were obvious differences in length, width and the ratio of the length to the width of leaves, the density and length of the non-glandular hair, the density and size of the stomata in the epidermis. [Conclusion] The morphological characters of the leaves mentioned above may serve as the identification standards of major cultivars of R. glutinosa.

  • Study on Quality Standard of Salvia miltiorrhiza Bunge Seedlings in Shandong Province

    [Objective] To preliminarily establish the quality classification standard of Salvia miltiorrhiza Bunge seedlings. [Method] 26 batches of S. miltiorrhiza seedlings were collected from Shandong Province in total. Plant height, number of inflorescence and branches, the yield were observed and compared. The data was analyzed by statistical software SPSS17.0. [Result] S. miltiorrhiza seedlings were divided into four levels. There were significant differences in the growth state, yields and quality among different grades. [Conclusion] The quality classification standard could be used for seedlings chosen of S. miltiorrhiza seedlings in production.

  • Study on Quality Standard of Hibiscus mutabilis L. Leaves

    [Objective] To establish the method for the quality control of Hibiscus mutabilis L. leaves. [Method] Microscopic identification and TLC were used to identify H. mutabilis leaves quantitatively. HPLC method was adopted to determine the content of active component rutin. [Result] Microscopic characteristics of H.mutabilis leaves were distinguishing. TLC spots were clear and well-separated. The linear range of rutin was 0.018 8-0.941 2 μg (r=0.999 6) with an average recovery rate of 97.97% (RSD=1.97%, n=6). The average content of rutin was 1.780 7 mg/g in 6 bathes of H.mutabilis leaves. [Conclusion] The established standard can be used to control the quality of H.mutabilis.

  • ICP-OES Determination of Inorganic Elements in Panax quinquefolium L.and Its Processed Products

    [Objective] To measure the content of 23 inorganic elements in Panax quinquefolium L. samples from different growth years, different harvesting periods, different medicinal parts, different commercial specifications and different processed products, including Al, B, Ba, Ca, Cd, Co, Cu, Fe, Mg, Mn, Mo, Na, Ni, P, Pb, Se, Si, Sn, Sr, Ti, V, Zn and K; To respectively count 6 trace elements that were essential to human body, including Zn, Cu, Fe, Se, Mo, Mn, and 2 toxic elements As and Hg; To investigate the medicinal value of P.quinquefolium medicinal materials from different growth years, different harvesting periods, different medicinal parts, different commercial specifications and different processed products. [Method]After P.quinquefolium medicinal materials had been digested by HNO3-H2O2, the content of all elements was measured using microwave assisted digestion-inductively coupled plasma-optical emission spectrometer (ICP-OES). [Result] The average recovery rate of all elements ranged from 92.7% to 106.1% with RSD below 4.5%. All elements did not interfere with each other under the optimized experimental conditions, so they could be determined at the same time. It was discovered by principal component analysis that Al, K, Ti, B, Sn, Cd, Na, Mo and Zn were the characteristic elements of P.quinquefolium samples. As for different commercial specifications, Fe and Mn contents were relatively higher, Zn content was at a moderate level and the Cu, Mo and Se content were relatively lower. The contents of inorganic elements were higher in three-year-old samples than in two-year-old and four-year-old samples. Moreover, the contents of inorganic elements in fibrous roots were higher than those in lateral roots. Different processed products of P.quinquefolium contained higher contents of Fe and Mn. Fe content of Red RADIX PANACIS QUINQUEFOLII was significantly higher than that of White RADIX PANACIS QUINQUEFOLII. There was little difference in Mn and Zn contents. Cu, Mo and Se contents were relatively lower in P.quinquefolium samples. [Conclusion] There were no obvious correlations between the contents of 6 essential elements and growth years, harvesting periods and medicinal parts. The roots, stems and leaves of P.quinquefolium contained higher contents of Ca, K, Mg and P, as well as lower contents of Mo, Sr, Ti, Co and Cd. Hg and other toxic metal elements were not found in P.quinquefolium samples. These pointed out that the roots, stems and leaves of P.quinquefolium were safe for human body with nutritional value and medicinal value. Therefore, we suggested developing stems and leaves of P.quinquefolium Red RADIX PANACIS QUINQUEFOLII into commodities for food, medicine and veterinary use every year. In addition, it is necessary to further study Red RADIX PANACIS QUINQUEFOLII, the processed product of P.quinquefolium.

  • Effects of Different Planting Densities on the Yield of Sinopodophyllum hexandrum Seedlings

    [Objective] To explore the change law of Sinopodophyllum hexandrum yield under different densities and seek the optimal planting density, so as to provide the scientific basis for forest-herb intercropping and GAP planting of S. hexandrum in fields. [Method] Under the same soil, fertilization and management method, S. hexandrum seedlings were planted under 12 different densities. [Result] The optimal planting density was 80 000-130 000 seedlings per hectare. There were obvious increases in the yield and income of S. hexandrum, that is, significant economic benefit when the plant spacing and line spacing were 25-30 and 30-35 cm, respectively. [Conclusion] It is appropriate to plant 80 000-130 000 S. hexandrum seedlings per hectare with the purpose of increasing both yield and income.

  • Photosynthetic Characteristics of Blumea balsamifera under the Condition of Shading Treatments

    [Objective] To study the photosynthetic characteristics of Blumea balsamifera (Linn.) DC under the condition of shading treatments. [Method] Under pot culture, B. balsamifera leaves with 1 (7%-10%), 2 (15%-20%), and 4 (30%-40%) layers of shading were studied. Li-6400 portable photosynthetic analyzer was used to detect and compare the photosynthesis daily change and the light response curve of B. balsamifera leaves. Photosynthetic characteristics of photosynthesis to the light intensity of B. balsamifera were studied. [Result] Daily change of photosynthetic rate (Pn) in one layer shading treatment showed a double-peak curve. The peak values were at 10:30 and 13:30 and the curve bottom was at 12:30, showing significant phenomenon of "midday depression". As for the 2- and 4-layer shading treatments, photosynthesis was relatively weak and was fluctuated around 0 in the whole daytime. Photosynthetic rates of B. balsamifera in three shading treatments were in the order of 1-layer shading > 2-layer shading > 4-layer shading. The responses of photosynthetic rate (Pn) were in the order of 1-layer shading > 2-layer shading > 4-layer shading. [Conclusion] The photosynthetic rate in 1-layer shading treatment was significantly higher than those in other two treatments, showing obviously high photosynthetic efficiency. Leaves had different photoresponse characteristics to different photosynthetic active radiations.

  • Effects of Different Zn Levels on the Growth, Nutrient Absorption and Effective Component Contents of Paris polyphylla var. yunnanensis

    [Objective] To research on the effects of Zn application on the output and effective component contents of Paris polyphylla var. yunnanensis. [Method] Under the condition of 70% plastic shed, pot experiment was carried out. There were in all 4 application levels of Zn, which were 0, 0.5, 1.0 and 2.0 mg/kg. Effects of Zn level on the growth, nutrient elements contents, and total saponins contents of rhizomes of Paris polyphylla var. yunnanensis. [Result] Compared with no application of Zn, Zn application significantly enhanced the plant height and total biomass of Paris polyphylla var. yunnanensis; but the increase of rhizomes was not significant. With the increase of Zn application, N contents in different parts firstly enhanced and then reduced. Zn application had synergistic effects on the P content in above ground stems, but had antagonistic effects on the P contents in leaves, new and old rhizomes. Zn application had synergistic effects on the K contents in leaves and new rhizomes but had antagonistic effects on the K content in stems. With the increase of Zn level, Zn content significantly enhanced in leaves and above ground stems, firstly increased and then reduced in new rhizomes; and there were no significant change laws in old rhizomes. With the increase of Zn levels, total saponins content of old and new rhizomes showed a downward trend in general. [Conclusion] Zn had certain effects on the output and effective components contents of Paris polyphylla var. yunnanensis; but its mechanism still needed further research.

  • Effects of Paeoniflorin on the Pharmacokinetics of Glycyrrhizic Acid and Glycyrrhetinic Acid in Rats

    [Objective] To explore the effects of paeoniflorin on the pharmacokinetics of glycyrrhizic acid and its active metabolite glycyrrhetinic acid after. [Method] After the administration of single glycyrrhizic acid or co-administration with paeoniflorin, blood samples were collected at different time points. The plasma concentration-time curves of glycyrrhizic acid and glycyrrhetinic acid were established. And pharmacokinetic parameters were calculated and analyzed by Topfit 2.0 software. [Result] Peak concentration (Cmax) and AUC of glycyrrhizic acid after co-administration with paeoniflorin reduced to 9% and 33% of those after administration of single glycyrrhizic acid, respectively. Tmax prolonged significantly; CL increased and Vd became larger. As for glycyrrhetinic acid, only the t1/2 prolonged significantly while other pharmacokinetic parameters had little changes. [Conclusion] Paeoniflorin dramatically inhibited the absorption rate and extent of glycyrrhizic acid in rats, but had only small effects on the absorption of glycyrrhetinic acid.

  • Determination of Polysaccharides in Ginseng Stem Leaves by Phenol-Sulfuric Acid Method

    [Objective] To optimize the factor conditions of the polysaccharides in ginseng stem leaves by phenol-sulfuric acid method. [Method] Polysaccharides contents in ginseng stem leaves were detected by UV Spectrophotometry; detection condition was optimized by orthogonal test. [Result] The optimal wavelength was 485 nm for the determination of polysaccharides contents in ginseng stem leaves by phenol-sulfuric acid method. The optimal color condition was A1B2C1, which was 1.0 ml of 6% phenol, 6.0 ml concentrated sulfuric acid, and 25 min reaction time. Crude polysaccharides had good linear relationship with absorbency between 10 and 70 μg/ml. The regression equation was Y=0.013 2X-0.085 7; correlation coefficient R was 0.999 6; recovery rate of test method was 100.36%, and RSD was 1.26%. The coloured solution had good stability within 120 min. [Conclusion] This method was accurate and reliable, and provided references for the further development and utilization of ginseng stem leaves.

  • Determination of Isofraxidin of Acanthopanax giraldii Harms by RP-HPLC

    [Objective] To establish an RP-HPLC method for determination of isofraxidin of Acanthopanax giraldii Harms and to compare the isofraxidin content of different medical parts, different producing areas and different collection time of A. giraldii Harms. [Method] The RP-HPLC analysis was carried out at 25 ℃ on a Kromasil C18 column. The mobile phase consisted of acetonitrile-methano1-0.1 % phosphoric acid (V/V/V, 20∶ 10∶ 70) and its flow rate was 1.0 ml/min. The detective wavelength was set to 343 nm. [Result] The calibration curve was linear within the concentration ranges of 0.60×10-3 to 7.49 μg/ml. The regression equation for isofraxidin was written as y=2.636×104x-1.051×103, (r=0.999 7). The average recovery was 99.15% with RSD of 0.95%. [Conclusion] This method was simple, accurate and replicate. And it can be adopted for the quality control of A. giraldii Harms.

  • Study on Volatile Compounds of Alpinia officinarum Hance

    [Objective] To analyze volatile compounds of Alpinia officinarum Hance. [Method] Simultaneous distillation and extraction (SDE)-GC-MS technology was set up to analyze the volatile compounds of A.officinarum. [Result] A total of 72 volatile compounds were isolated and identified from A.officinarum, the main ones were 1, 8-cineole (38.54%), α-terpinenyl acetate (6.55%), torreyol (7.74%), isolongifolene (3.64%), ledene (3.93%) etc. [Conclusion] The information about volatile compounds of A.officinarum obtained by SDE is different from those obtained by steam distillation or headspace analysis, and more plentiful than them. We can get a better representation system for the volatile compounds of A.officinarum when combine these methods together.

  • Aroma Chemical Components from the Pine Needles of Pinus massoniana Lamb

    [Objective] To research on the aroma chemical components from the pine needles of Pinus massoniana Lamb. [Method] Ethanol extraction and acetic ether extraction were adopted. Based on silica gel column chromatography separation, components of pine needles were identified and analyzed by GC-MS, HPLC and standard comparison. [Result] A total of 6 components were isolated from the pine needles. And the main components were vanillin and dihydroactinidiolide. [Conclusion] The two components were isolated from the pine needles of P. massoniana for the first time.

  • Determination of Aradecoside A Contents in Aralia Plants in Different Production Areas by HPLC

    [Objective] To detect the aradecoside A contents in Aralia plants in different production areas by HPLC. [Method] Chromatographic column was Kromasil C18 column (250 mm × 4.6 mm, 5 μm). Mobile phase was acetonitrile-0.05% phosphoric acid solution (V/V, 35∶65); detection wavelength was 205 nm; column temperature was 30 ℃; flow rate was 0.8 ml/min; injection volume was 20 μl. [Result] Aradecoside A had good linear relationship with peak area between 0.632 5-25.30 μg/ml (r=0.999 9). The average recovery rate was 97.97% and RSD was 1.02%. [Conclusion] This method was accurate, sensitive, and repeatable and could be used for the quality control of root barks of Aralia plants.

  • Effects of Growth Years on the Flavoniods Content of RADIX OPHIOPOGONIS

    [Objective] To research the effects of growth period on the flavoniods content of RADIX OPHIOPOGONIS, and to provide scientific references for the GAP quality control of RADIX OPHIOPOGONIS and development and utilization RADIX OPHIOPOGONIS resources. [Method] Production area of three-year-old RADIX OPHIOPOGONIS in Hangzhou City was studied by in-situ dynamic method. Random collection of Ophiopogon japonicus root tuber was conducted after 0.5, 1, 2 and 3 years of cultivation. After washing, the O. japonicus fresh root tuber was dried at -50 ℃. Flavoniods contents of O. japonicus dry root tubers (RADIX OPHIOPOGONIS) under different treatments were detected by UV spectrophotometry. Correlation rules of the flavoniods contents in RADIX OPHIOPOGONIS with different growth years were analyzed by SPSS17.0 software. [Result] The average content of flavoniods in RADIX OPHIOPOGONIS was between 17.68% and 34.86%. With the increase of RADIX OPHIOPOGONIS growth years, flavoniods content showed extremely significant positive increase (P<0.001). The regression equation was y1=-0.008x2+0.066x+0.046 (R2=0.999), and y2=-5E-08x2+0.047 (R2=0.998). [Conclusion] Growth years of RADIX OPHIOPOGONIS had close correlation with the content of flavoniods. There was scientific basis for the collection of three-year-old O. japonicus in main production areas. Due to the particular climate in the main production areas, root tubers should be collected at one year old; and they were easily decayed in the summer at the second year due to the high temperature and humidity.

  • Degradation Kinetics of Scutellarin in Water Extract of HERBA SCUTELLARIAE BARBATAE

    [Objective] To study the change of scutellarin in HERBA SCUTELLARIAE BARBATAE under different immersing conditions. [Method] HPLC was adopted to detect the content of scutellarin in water extract of HERBA SCUTELLARIAE BARBATAE immersed at different temperatures and times. The scutellarin content and soaking time were selected as variants of dynamic equation and the kinetics parameters were calculated. [Result] Under the immersing conditions of temperature from 20 to 60 ℃,the change of scutellarin content tended to show degradation characteristic in water extract of HERBA SCUTELLARIAE BARBATAE, and degradation rate was the fastest between 30 and 40 ℃. While the scutellarin content in water extract remained unchanged at the immersing temperature above 70 ℃ and it had a higher extraction rate. [Conclusion] Scutellarin of HERBA SCUTELLARIAE BARBATAE exists enzymatic degradation reaction in the process of immersion. It is better to put HERBA SCUTELLARIAE BARBATAE at the temperature above 70 ℃ when water extracting, in which way can effectively increase the extraction rate of scutellarin.

  • Study on Stability and Cutaneous Penetration Kinetics in Vitro of Cationic Liposome Gels Containing Paeonol

    [Objective] To prepare cationic liposome gels containing paeonol, and to study its stability and cutaneous penetration kinetics in vitro. [Method] To prepare the liposome gel by dispersion-ultrasonic and gridding method, and study the stability with the impact factor experiments, and compare the penetration rate of liposome gel with conventional gel in vitro using the Franz-diffusion cell. [Result] Mean diameter of the liposome was (132.7±14.1) nm with Zeta potential of (+33.54±1.95) mV and mean entrapment efficiency of (73.04±1.24)% (n=3). Drug content of the liposome was (3.17±0.13) mg/g (n=3). The liposome gel had a promising appearance. It was stable at the humidity and the room temperature while was sensitive at the light and the temperature from 40 to 60 ℃. The cumulative penetration amount of the liposome gel was higher than that of the conventional gel (P<0.05). Its cutaneous penetration rate and cumulative amounts in skin were higher than those of the conventional gel (P<0.05). [Conclusion] The cationic liposome gels containing paeonol was stable and feasibly prepared, and enhanced the cutaneous penetration efficiency and guaranteed the persistent release rate.

  • Isolation, Purification and Activity Analysis of Polysaccharides from RHIZOMA ATRACTYLODIS MACROCEPHALAE

    [Objective] To extract and purify the polysaccharides from RHIZOMA ATRACTYLODIS MACROCEPHALAE; and to discuss the effects of polysaccharides on lymphocyte transformation of mice spleen lymphocyte. [Method] Polysaccharides (cPAM, zPAM, PAM) were extracted and isolated from RHIZOMA ATRACTYLODIS MACROCEPHALAE by alcohol precipitation method, deproteinization, and Sephadex G-75 preparative column. Lymphocyte transformation in vitro of the three kinds of polysaccharides was researched. [Result] The three kinds of polysaccharides all had significant promotion effects on the lymphocyte proliferation of mice spleen activated by ConA. 5 μg/ml zPAM showed significant promotion effects on the proliferation of mice spleen lymphocyte; and 5 and 10 μg/ml PAM also had significant promotion effects on the proliferation of mice spleen lymphocyte. [Conclusion] The purification degree of RHIZOMA ATRACTYLODIS MACROCEPHALAE polysaccharides had certain relationship with the proliferation of mice spleen lymphocyte. Polysaccharides with high purification degree could promote the proliferation of mice spleen lymphocyte. Crude polysaccharides from RHIZOMA ATRACTYLODIS MACROCEPHALAE had no effects on the proliferation of mice spleen lymphocyte.

  • Study on Bacteriostasis and Anticancer Activity in Vitro of Potentilla anserine L. Extracts

    [Objective] To investigate the bacteriostasis and anticancer activity in vitro of Potentilla anserine L. extracts in different solvents. [Method] The four different kinds of solvents (petroleum ether, ethyl acetate, ethanol and water) were used to extract P. anserine root tuber in turn, so as to obtain corresponding extracts. Antibacterial activities of the four extracts for the four kinds of bacteria (Escherichia coli., Salmonella sp., Staphylococcus aureus, and Bacillus subtilis) were observed by using Oxford cup double-plate method. Besides, the inhibitory effects of the extracts on human hepatoma cell HepG2 were studied in vitro by crystal violet staining. [Result] Among the four extracts, ethanol had the strongest antibacterial activity and the most obvious anti-cancer cell activity; and its IC50 was 375 μg/ml. [Conclusion] There were antibacterial and anti-cancer compounds in P. anserine. The inhibitory activity of ethanol extracts was the most remarkable among the four different solvents.

  • Effects of Different Hormones on the Suspension Cell Growth and Development of Bupleurum falcatum L.

    [Objective] To study the effects of different hormones the suspension cell growth and development of Bupleurum falcatum L.. [Method] The plant growth hormone of medium for Bupleurum falcatum L. suspension cell cultures was optimized by the test method of uniform design. [Result] The optimal hormone formulation of B. falcatum suspension cell cultures was N6 + 0.5 mg/L NAA + 0.75 mg/L 6-BA + 1.5 mg/L KT. Based on the optimal hormone formulation to fit the curves about the relationship of the cell fresh-weight t and the cell dry-weight in incubation time, the fitting equation of the relationship between the cell fresh-weight and the incubation time was Y=5.703 6X2-41.429X+136.44; the fitting equation of the relationship between the cell dry-weight and the incubation time was Y=2.021 8X2-22.595X+57.19. [Conclusion] This study can lay the foundation for the further study and the large-scale culture about the B. falcatum.

  • Optimization of Culture Conditions of Se-enriched Ganoderma lucidum (Leyss.ex Fr.) Karst.

    [Objective] To find the optimal culture conditions of Se-enriched Ganoderma lucidum (Leyss.ex Fr.) Karst. mycelium. [Method] The effects of carbon source, nitrogen source and fermentation conditions on the dry weight, total selenium and organic selenium content of Se-enriched G. lucidum mycelium were studied by using single factor test and orthogonal test. [Result] Under conditions of 0.01% sodium selenium content, glucose content 4%, potato extracting 20%, peptone content 1%, KH2PO4 0.3%, MgSO4 0.15%, pH=7.0, inoculation amount 10%, culture temperature 28 ℃, and shaking speed 180 r/min, the dry weight of Se-enriched G. lucidum mycelium was 15.28 mg/ml, and the organic selenium content was 4 115.36 μg/g. [Conclusion] The optimal culture conditions of Se-enriched G. lucidum were determined, which could provide basis for development and utilization of Se-enriched G. lucidum.

  • Study on Enzyme-assisted Extraction of Indirubin from Baphicacanthus cusia (Nees) Bremek Leaves

    [Objective] To observe the key parameters of enzymatic extraction that impact extraction of indirubin from Baphicacanthus cusia (Nees) Bremek leaves. [Method] The content of indirubin was determined using HPLC. Effects of enzyme dosage, hydrolysis pH and hydrolysis time on extraction rate of indirubin were investigated with the aim of optimizing its extraction technology. [Result] The optimal enzyme-assisted extraction conditions of indirubin from B. cusia leaves were the combination of enzyme dosage 25 U/g, hydrolysis pH 4.8 and hydrolysis time 6 h. Under the above conditions, the Z value of indirubin was 7.02%. [Conclusion] Enzyme-assisted extraction method is suitable for extraction of indirubin from B. cusia leaves.

  • Research Status and Progress of the Triterpenoids in Ganoderma lucidum

    The extraction and purification technologies of triterpenoids in Ganoderma lucidum were reviewed, as well as the research progress of chemical component and content analyses. Triterpenoid was an important active pharmaceutical ingredient in G. lucidum. However, due to the low content and complex extraction and purification processes, only a small amount of high-purity G. lucidum triterpenoids were obtained; and it was still at the status of laboratory research. Based on these, industrialization technology of the preparation of G. lucidum triterpenoids was further discussed, aiming at promoting the rapid development of G. lucidum industry, and providing the scientific references for the research and production of relevant drugs and health foods.

  • Analysis of the Nutritional Components of Physalis alkekengil var. francheti and P. pubescens L.

    [Objective] To research the nutritional components of Physalis alkekengil var. francheti and P. pubescens L. [Method] Water content, ash content and crude fat content were detected by Soxhlet extraction. Protein content was detected by Kjeldahl Method. Inorganic elements were analyzed by the detection method of GB/T12393-90. Fat-soluble vitamins and water-soluble vitamins were detected by GB 12388-90 and GB 12392-90, respectively. [Result] There were abundant inorganic elements in the fruits and persistent calyx of P. alkekengi and P. pubescens. Among them, K, P, Na, Mg and Ca had relatively high contents; while Pb and As were not detected. Amino acids were abundant, which had more than 18 kinds. Among them, aspartic acid had the maximum content. Besides, there were abundant vitamins. [Conclusion] Contents of inorganic elements and amino acids in persistent calyx were higher than those in fruits, which provided theoretical basis for the rational utilization of persistent persistent calyx.

  • Antioxidant Activity in Vitro of Flavonoids in RHIZOMA CYPER

    [Objective] To study the antioxidant effects of flavonoids in RHIZOMA CYPER. [Method] Taking 95% ethanol as the solvent, flavonoids were extracted from RHIZOMA CYPER by water bath reflux. Then, reduction ability and scavenging effect of RHIZOMA CYPER flavonoids on three free radicals (DPPH·, ·OH and O-2·) were researched by colorimetric method. [Result] Flavonoids in RHIZOMA CYPER had significant scavenging effects on the three free radicals; and their scavenging capabilities were in the order of DPPH· >·OH > O-2·. There were significant dose-effect relationship between the concentration and the scavenging rate. When the flavonoids concentration was 0.4 mg/ L, the scavenging rates to O-2·, DPPH· and ·OH were 25.5%, 67.0% and 28.5%, respectively. Fe3+ reducing ability enhanced as the concentration increased. [Conclusion] Flavonoids in RHIZOMA CYPER had relatively strong antioxidant activity.

  • Removal Processes of Acerbity and Dumb Tastes from Colocasia esculenta (L.) Schott Stems and the Determination of Amino Acids and Alkaloid Contents

    [Objective] To research the optimal methods for acerbity and dumb tastes removal from Colocasia esculenta (L.) Schott stems. [Method] Components with acerbity and dumb tastes in C. esculenta stems were processed by water boiling, ethanol and organic acid soaking, CO2, and salting. [Result] Calcium oxalate crystal, 3, 4-dihydroxybenzaidehyde and its glycoside in C. esculenta stems were the main irritant components in C. esculenta stems. They could be removed in different degress by the methods of boiling, ethanol, limewater soaking, salting and so on. [Conclusion] The acerbity and dumb tastes in C. esculenta stems were related to the calcium oxalate crystal, 3, 4-dihydroxybenzaidehyde and its glycoside. After long-time boiling, salting and the ethanol, organic acid and limewater soaking, the acerbity and dumb tastes reduced in different degrees. Among them, salting had the most significant effects in removing the acerbity and dumb tastes from C. esculenta stems. And verification test was conducted by HPLC.

  • Comparative Study on Extraction of Alkaloids from DATURAE FLOS by Different Methods

    [Objective] To compare the yield of effective parts, effective components and chemical composition from DATURAE FLOS extracts by microwave-assisted extraction (MAE), soaking extraction (SE) and backflow extraction (BE), so as to search the optimal extraction method. [Method] UV and HPLC were adopted to detect the content of total alkaloids, scopolamine and atropine in ethanol extracts of DATURAE FLOS by MAE, SE and BE. And their chemical constituents were investigated by TLC. [Result] The extraction yield of total alkaloids by MAE, BE and SE were 0.42%, 0.38% and 0.32%, respectively. The extraction yield of scopolamine by MAE, BE and SE were 0.24%, 0.24% and 0.13%, respectively. The extraction yield of atropine by MAE, BE and SE were 0.15%, 0.10% and 0.08%, respectively. It was discovered by TLC comparison that there was no significant difference in spots of alkaloids among three methods, that is, all of them showed four similar spots of alkaloids. [Conclusion] Characterized by short time, less solvent consumption, simple operation and high efficiency, MAE can be use to extract alkaloids from DATURAE FLOS. MAE is an environmental and energy-saving new technology which is worthy to be popularized and applied in large scale extraction of alkaloids from DATURAE FLOS.

  • Component Analysis of Aqueous Extracts from FRUCTUS LIGUSTRI LUCIDI and the Content Determination of Its Polysaccharides

    [Objective] To analyze the components of aqueous extracts from FRUCTUS LIGUSTRI LUCIDI to detect the content of polysaccharides. [Method] Components of aqueous extracts were extracted by decocting method. The components were analyzed and identified by chemical method. Polysaccharides content in aqueous extracts was detected by phenol-sulfuric acid method. [Result] There were various components in FRUCTUS LIGUSTRI LUCIDI aqueous extracts, such as water-soluble protein, triterpenoid saponins, flavonoids and their glycosides, dihydro-anthraquinones, volatile oils, polysaccharides, and polyphenols. Polysaccharides content in FRUCTUS LIGUSTRI LUCIDI reached 10.3 mg/g. [Conclusion] There were various types of the components of aqueous extracts from FRUCTUS LIGUSTRI LUCIDI; and the content of polysaccharides was relatively high.

  • Research on Sustainable Utilization of Traditional Chinese Medicine Resources in Enshi Prefecture

    An investigation on the current situation and utilization of medicinal plant resources in Enshi Prefecture, Hubei Province is summarized, and studies on the resource protection and culture of important medicinal plant germplasm resources were concluded. The research direction of sustainable utilization of traditional Chinese medicine resources in Enshi Prefecture was put forward∶ speeding up the establishment of resource conservation system, improving the breeding and replacement of endangered medicinal plants; actively developing the resource investigation of medicinal plants, setting up dynamic monitoring and forecasting net work system for medicinal plant resources; strengthening the research of Chinese medicinal materials resource ecology, promoting the selection and breeding research; promoting the research of Chinese medicine material GAP, enhancing the research of exploitation and utilization in an all-round way; exploring the application of DNA barcoding in plants, providing new ideas for original plant identification, endangered species protection and resource evaluation of medicinal plants.

  • Research Progress of Ganoderma sinense

    In order to protect the protect the unique germplasm resources of China and to promote the development of G. sinense industry, the biological characteristics, active nutritional compositions, pharmacological action, production and industrialization of G. sinense mycelium, fruiting body and variety were reviewed in this research based on relevant literatures. The main problems in the research of G. sinense were discussed. There were few reports on the investigation and collection of G. sinense germplasm resources and the breeding of high-quality strains. In-depth researches on the cultivation and fermentation technologies of G. sinense were lacked. Based on these, researches on G. sinense should be carried out in the aspects of germplasm resources, strain breeding, production technology, active component analysis, and pharmacological efficacy components, so as to protect the unique germplasm resources of China and to promote the development of G. sinense industry.

  • Comparison of the Cold-resistance Capabilities of Four Ligustrum Cultivars

    [Objective] To compare the cold-resistance capabilities of four Ligustrum cultivars. [Method] Based on semilethal low temperature test and low temperature stress test in phytotron, cold-resistance capabilities of two-year-old seedlings of 4 Ligustrum cultivars were compared at leaf-expansion and shoot-growing stage. [Result] At leaf-expansion and shoot-growing stage, the semilethal low temperatures of Ligustrum×Vicaryi Hort., Ligustrum lucidum Ait., Ligustrum japonicum Thunb. 'Howardii' and Ligustrum lucidum Ait. 'Excelsum Superbum' were -6.34, -5.69, -4.55 and -3.27 ℃, respectively. SOD activity, soluble protein content, soluble sugar content and MDA content in leaves under low temperature stress were all higher than those under normal temperature. As the low temperature stress enhanced, soluble protein content in leaves firstly enhanced and then decreased. Among the 4 cultivars, soluble sugar content in 3 cultivars firstly increased and then decreased; and MDA content continuously enhanced. [Conclusion] The cold-resistance capabilities of Ligustrum×Vicaryi Hort. and Ligustrum lucidum Ait. were higher than other cultivars; Ligustrum japonicum Thunb. 'Howardii' was middling; and Ligustrum lucidum Ait. 'Excelsum Superbum' was the worst.

  • Determination of IC_(50) for Nine Medicinal Plants against α-Glucosidase Activity

    [Objective] To screen the plant-derived α-glucosidase inhibitors. [Method] Using the half maximal inhibitory concentration (IC50) as the potency index, the inhibitory effects of water extracts from 34 kinds of traditional Chinese medicines on α-glucosidase activity were evaluated. [Result] When they were used in concentrations of 50 mg/ml, the water extracts from RADIX MORINDAE OFFICINALIS, RADIX ACHYRANTHIS BIDENTATAE, RHIZOMA POLYGONATI, RADIX PAEONIAE ALBA, RADIX POLYGONI MULTIFLORI, FRUCTUS SCHISANDRAE CHINENSIS, RHIZOMA CHUANXIONG, RHIZOMA ET RADIX POLYGONI CUSPIDATI, RHIZOMA DIOSCOREAE NIPPONICAE showed inhibitory effects on α-glucosidase activity with inhibition rate over 50%. Further researches found that the inhibition rate of water extracts from FRUCTUS SCHISANDRAE CHINENSIS, RHIZOMA CHUANXIONG and RHIZOMA ET RADIX POLYGONI CUSPIDATI on α-glucosidase activity was all higher than 80%. The above 3 kinds of traditional Chinese medicines were ranked in descending order of inhibitory effects: RHIZOMA ET RADIX POLYGONI CUSPIDATI > FRUCTUS SCHISANDRAE CHINENSIS > RHIZOMA CHUANXIONG. Among them, there were 3 medicinal plants with IC50 below 40, which were arranged in order of efficiency strength, RADIX POLYGONI MULTIFLORI > RADIX PAEONIAE ALBA > RHIZOMA ET RADIX POLYGONI CUSPIDATI. [Conclusion] It is feasible to screen hypoglycemic drugs from plant resources. This study can provide reference for discovering high-efficiency and safe biological drugs of reducing postprandial blood glucose.
