  • Reseach on Quality Classification Standard of Fritillaria cirrhosa D. Don Seed

    [Objective]To understand the morphological characteristics and germination rate of Fritillaria cirrhosa D. Don seeds and provide the basis for seed quality standard.[Method] The purity,1 000-grain weight,water content,water absorption rate and germination rate of Fritillaria cirrhosa D. Don seed were determined,to establish quality classification standard of Fritillaria cirrhosa D. Don seed. [Results] The purity was 92. 48% ,and the germination power was 60. 45% ,and the germination rate was 94. 80% ,and the water content was 13. 81% , and the maximum water absorption rate was 160. 18% ,and the life force was 90. 40% ,and the 1 000-grain weight was 1. 724 g,disease was 3. 748 0% ,dead insects and eggs was 0. 030 8% . [Conclusion] The quality classification standard of Fritillaria cirrhosa D. Don seed was established to the basis for the artificial cultivation of Fritillaria cirrhosa D. Don.

  • Effect of the Jobstears Planting Pattern on Its Growth and Yield

    [Objective]The effect of jobstears planting pattern on its growth and yield was explored for the theoretical basis of its high-efficiency,high-yield and good quality cultivation. [Method]The field experiment in 6 different jobstears cultivation patterns was conducted for the comparison with the jobstears planting done by farmers themselves(as CK) . [Results] The statistical analysis showed that the different cultivation methods greatly impacted on the jobstears growth and yield. From the experimental results in the cultivation,the best planting pattern was that the planting distance of plant was 30 cm × 80 cm (with 4. 168 5 million holes/hm2) ,which yield was the increment of 37.41% compared with that of CK. Next one was 60 cm × 60 cm under plastic film covering,which yield was the increment of 31. 65% compared with that of CK.

  • Determination of β-sitosterol Content in Different Species of HERBA MOGHANIAE by RP-HPLC-ELSD

    [Objective]To detect the contents of β-sitosterol in the roots and stems of Moghani glutiaosa (Prain) Y. T. Wei and M. latifolia Benth. by RP-HPLC-ELSD,as well as the roots of M. philippinensis (Merr. et Rolfe) Li and M. macrophylla (Willd.) O. Kuntze in different grown areas. [Methods] Chromatographic column was Kromasil C18 (250 mm × 4.6 mm,5 μm) ; column temperature was 25 ℃; flow rate was 1. 0 mL/min; mobile phase was methanol. [Results] β-sitosterol showed good linear relationship between 0. 000 65 and 0. 003 25 μg/mL, the average recovery rate was 99. 4% (n = 5) and RSD was 0. 87% . β-sitosterol contents in M. glutiaosa and M. latifolia roots and stems were both relatively high. M. philippinensis grown in Yichang City of Hubei Province had the highest content of β-sitosterol; while M. macro-phylla grown in Huaxi Botanical Garden had the highest content of β-sitosterol. β-sitosterol contents in other grown areas were relatively low. β-sitosterol content in the roots of M. macrophylla was higher than that in M. philippinensis. [Conclusions]This method was simple,rapid and accurate and could be used for the quality control of RADIX MOGHANIAE.

  • Composition and Detection of Monosaccharide in the Polysaccharides of Juglans sigillata D. Shells

    [Objective]To establish the quantitative detection method for the monosaccharide composition in the polysaccharides of Juglans sigillata D. shells. [Methods]Using trifluoroacetic acid to hydrolyze the polysaccharides of J. sigillata shells,1-phenyl-3-methyl-5-pyrazolone (PMP) was added into the hydrolysate of polysaccharides for pre-column derivatization. PMP derivative of monosaccharide was analyzed by high performance liquid chromatography (HPLC) . Chromatographic column was Agilent Venusil XBP-C18 (4.6 mm × 250 mm,5 μm) ; mobile phase was acetonitrile solution ammonium acetate buffer (pH 5. 5,19∶ 81,V/V) ; detection wavelength was 250 nm; column temperature was 30 ℃ ; and volume flow was 1 mL/min. [Results] Polysaccharides of J. sigillata shells were composed of 11 monosaccharides,which were D-glucuronic acid,D-mannose,galactose,D-arabinose,rhamnose,D-galacturonic acid,glucose and other 4 monosaccharides not detected. The linear ranges of D-mannose,rhamnose,D-glucuronic acid,D-galacturonic acid,glucose,galactose and D-arabinose were 0. 041 8-2. 09,0. 045 9 2. 295,0. 043 5 2. 175,0. 042 7 2. 135,0. 040 5 2. 026,0. 043 9 2. 195 and 0. 047 5 2. 375 μg/mL,respectively. And their average recovery rates were 95. 9% ,103. 1% ,104. 6% ,97. 5% ,95. 8% ,87. 5% and 98. 1% ,respectively. The RSD values were all smaller than 3. 0% . Among them,D-arabinose had relatively high average content. [Conclusions]This method could accurately detect the monosaccharides and their contents in the polysaccharides of J. sigillata shells.

  • Optimization of Extraction Technology of Quercetin in Fagopyrum tataricum (L.) Gaertn. Bran by Orthogonal Method

    [Objective]To optimize the extraction technology of quercetin in Fagopyrum tataricum (L.) Gaertn. bran. [Method]After the pretreatment of Fagopyrum tataricum (L.) Gaertn. bran,whose quercetin glycoside was enzymically degraded into quercetin,the further extraction was operated to study the effects of methanol concentration,solid-liquid ratio and extraction temperature on the extraction yields of quercetin in Fagopyrum tataricum (L.) Gaertn. bran. [Results] In the test range,with the increasing of methanol concentration,temperature and solid-liquid ratio,the quercetin extraction rate of Fagopyrum tataricum (L.) Gaertn. bran increased gradually; the sequence of factors affecting extraction rate was methanol concentration,temperature and solid-liquid ratio; the optimal combination of factors was A4B4C4 , considering many aspects as the cost,workload and efficiency,and the combination of A3B2C3 was chosen under the optimal conditions which was extraction temperature of 35 ℃ ,ratio of solid to liquid of 1∶ 25 (g/mL) ,methanol concentration of 70% ,extraction time of 1 h. [Conclusion]After further testing,the quercetin extraction rate could reach 89. 3% under the optimal conditions with good reproducibility.

  • Effects of Harvest Time on the 5-HMF Contents in FRUCTUS LIGUSTRI LUCIDI and Its Processed Products

    [Objective] To detect the 5-HMF contents in FRUCTUS LIGUSTRI LUCIDI before and after stewed with wine. [Methods] FRUCTUS LIGUSTRI LUCIDI was harvested from August to December,and then was processed according to the method in Pharmacopoeia of People's Republic of China (edition 2010) . 5-HMF content in FRUCTUS LIGUSTRI LUCIDI was detected by HPLC. Chromatographic column was Inertsil ODS-3 C18 (4.6 mm × 250 mm,5 μm) ; mobile phase was acetonitrile water (10∶90,V/V) ; flow rate was 1 mL/min; detection wavelength was 285 nm; detection temperature was room temperature; and injection volume was 20 μL. [Results] 5-HMF content in FRUCTUS LIGUSTRI LUCIDI was the highest when harvested on August; and 5-HMF content in FRUCTUS LIGUSTRI LUCIDI stewed with wine enhanced by 12. 50 45. 95 times. [Conclusions] In order to effectively utilize the 5-HMF in FRUCTUS LIGUSTRI LUCIDI,timely harvest and processing were needed according to the actual needs.

  • The Processing Technology of Honey-fried FLOS FARFARAE

    [Objective]To optimize the processing technology of honey-fried FLOS FARFARAE. [Methods] L9 (34) orthogonal test was adopted. Honey dosage,moistening time,processing time and processing temperature were taken as the investigation factors. With the properties,the mass fractions of tussilagone and ethanol-soluble extractives as the comprehensive evaluation indices,the honey-fried processing technology of FLOS FARFARAE was selected. [Results]The optimal processing technology was 40% honey,4 h moistening time,6 min processing time and 100 110 ℃ processing temperature. [Conclusions] The optimal processing technology was stable and feasible,and could be used for the research on quality standards of FLOS FARFARAE.

  • Hormone Optimization and Callus Induction from the Protocorm of Dendrobium candidum Wall

    [Objective]To establish the tissue culture system for budlets induced from the protocorm callus of Dendrobium candidum Wall. ex Lindl. ,and to optimize the hormone in culture medium. [Methods]Budlets were induced from the callus of D. candidum under three culture conditions in culture mediums with eight concentrations of 2,4-D. [Results] During callus induction,the optimal culture medium was 0. 5 mg/L 6-BA + 0. 5 mg/L NAA + (1. 5-2. 5) mg/L 2,4-D + 20. 0 g/L sucrose + 200. 0 g/L mashed potato + 2. 0 g/L activated carbon + 10. 0 g/L agar. And callus induction had the optimal effects under the culture condition of illumination after dark culture for 15 d. [Conclusions]The budlets were the maximum under the optimal culture condition mentioned above.

  • Anti-hepatic Fibrosis Action of Perfoliate Knotweed Herb against Rat and Its Mechanism

    [Objective] To reveal the anti-hepatic fibrosis action of Perfoliate Knotweed Herb against N-Nitrosodimethylamine-induced rat model and its mechanism. [Method] Eighty SD rats were randomly divided into 6 groups: normal group,model group,Perfoliate Knotweed Herb medication groups(high,medium and low doses) and colchicine group. Except normal group,rats in all other groups were medicated by introperitoneal injection with 0. 5% DMN solution (1. 6 mL/kg) for continuous 3 d within a week. First week used 2 /3 dose and then full dose was adopted. colchicine medication group adopted 0. 1 mg/kg colchicine; high,medium and low doses of Perfoliate Knotweed Herb medication groups applied 7. 5,5. 0 and 2. 5 g/kg. Medication was performed once a day and continued for 4 weeks. At the end of 4th week,blood collected from eyeball execution was used for detection of serum hyaluronic acid (HA) ,Laminin(LN) ,precollagen III (PCIII) ,collagen IV (IV-C) by using ELISA. HF formation was observed by hematoxylin-eosin stain(HE stain) ; immunohistochemical assay was employed to detect the transformation growth factor β1 (TGF-β1) . [Results]In comparison with model group,serum HA,LN,PCIII and IV-C expression levels in high and medium dose Perfoliate Knotweed Herb medication groups were significantly lowered (P < 0. 05 or P < 0. 01) ; immunohistochemical assay indicated that TGF-β 1 level in high and medium dose groups were remarkably lowered (P < 0. 05 or P < 0. 01) ,while that in low dose group did not (P > 0. 05) . [Conclusion] Perfoliate Knotweed Herb is endowed with good efficacy on the treatment of N-Nitrosodimethylamine-induced hepatic fibrosis in rats,with the mechanism of down-regulating the expression of TGF-β1 ,thus inhibition the initiation of hepatic fibrosis.

  • Extraction Technology of Flavonoids Components from RADIX MOGHANIAE

    [Objective]To select the optimal extraction technology of flavonoids components from RADIX MOGHANIAE. [Methods] RP-HPLC method was adopted for the screening. Chromatographic column was Diamonsil C18 (250 mm × 4.6 mm,5 μm) ; mobile phase was anhydrous methanol 0. 5% phosphoric acid,0 →10 min(23∶77,V/V) ,11→55 min (43∶57,V/V) ; flow rate was 1 mL/min; column temperature was 30 ℃ ; and detection wavelength was 240 nm. [Results] The optimal extraction technology of total flavonoids from RADIX MOGHANIAE was as follows: 1∶16 solid-liquid ratio,80 ℃ reflux extraction for 2 h,and 3 extraction times.[Conclusions]This research provided references for the further research and extraction of RADIX MOGHANIAE.

  • Enrichment Technology of Total Flavonoids of Hawthorn Leaves by Macroporous Resin Adsorption Process

    [Objective]To research the technological conditions and parameters of the enrichment of total flavonoids of hawthorn leaves by macroporous resin adsorption process. [Methods]With the total flavonoids of hawthorn leaves as the investigation index,AB-8,D-101,DA-201,NKA-9 and DM-301 macroporous resins were selected. Column chromatography of the extracting solution of hawthorn leaves was carried out. The optimal technological condition for the enrichment of total flavonoids in macroporous resin was investigated. [Results] The optimal condition for D-101 macroporous resin to enrich the total flavonoids of hawthorn leaves was as follows: 0. 2 g/mL injection concentration of the extracting solution of hawthorn leaves,6 BV sample volume,and 1. 0 mL/min flow rate. Before desorption,4 BV distilled water was used for cleaning; and then desorption was carried out by 4 BV of 70% ethanol. Eluent was collected,so that the extracts of the total flavonoids in hawthorn leaves were obtained. The enrichment rate of total flavonoids reached 61. 85% . [Conclusions] This method could enrich the total flavonoids of hawthorn leaves. It was simple and feasible,and could meet requirements of industrialized production.

  • Determination of the Content of the Metal Cations in Polygonum aviculare by Highperformance Capillary Electrophoresis

    [Objective]To establish a method for determination of the content of the metal cations in Polygonum aviculare. [Methods]High-performance capillary electrophoresis was used,with uncoated fused-silica capillary (50μm i. d. × 50 cm,46 cm to detector) ,and the running buffer was 7 mmol/L imidazole and 6 mmol/L tartaric acid (pH = 6. 0) ,and the separation voltage was 20 kV,and the electrokinetic injection time was 3 s at the voltage of 15 kV,and the detection wavelength was 214 nm. [Results] The concentration of K+ ,Na+ ,Mg2+ , Mn2 + ,Ca2+ and Zn2+ showed a good linear relationship with their peak height in the range of 0. 5 120 mg/L (r = 0. 999 3) ,0. 5 120 mg/L (r =0.998 7) ,0.1 -60 mg/L (r =0.997 3) ,0. 1 -120 mg/L (r =0. 999 2) ,0. 1-60 mg/L (r =0. 999 1) and 0. 5-120 mg/L (r = 0. 998 2) ; the average recovery of K+ ,Na+ ,Mg2 + ,Ca2+ ,Zn2+ were 103. 5% ,96% ,93. 4% ,105. 4% and 101. 6% ,and the RSD were 1. 75% ,1. 66% ,1. 52% ,1. 43% and 1. 19% (n = 6) . [Conclusion]The method is simple,rapid,accurate,reliable,and can be used for determination of metal cations content in Polygonum aviculare.

  • The Limit Detection Method of the Ginkgoic Acid in the Extracts from the Leaves of Ginkgo biloba L.

    [Objective]To compare the limit detection methods of ginkgoic acid in the extracts from the leaves of Ginkgo biloba L. by Chinese Pharmacopoeia (2010 edition) and European Pharmacopoeia (EP7. 2) ; and to analyze and evaluate the preparation methods,chromatographic conditions and calculation methods of the two samples. [Methods]10 batches of the extracts from G. biloba leaves were prepared into tested substances by the two standards. Crossover trial was carried out; and the content of ginkgoic acid (GA) was calculated. [Results]By using the same detection and calculation method,GA content in the tested solution by EP7. 2 method was 5. 62 times of that by Ch. P2010 method. By using the same preparation method,GA content in the tested solution by Ch. P2010 standard was 1. 43 times of that by EP7. 2 standard. [Conclusions]The detection results of the two standards had poor regularities; each standard had their own advantages and disadvantages. Therefore,it was necessary to combine the advantages of the two to establish an accurate and reliable limit detection method for GA.

  • Anti-aging Effects of the Flavonoids of Oxytropis glabra DC. on Mice

    [Objective]To discuss the anti-aging effects of the flavonoids of Oxytropis glabra DC. on the mice of senile model. [Methods] D-galactose was used to prepare the senile mice model. Then,mice were given the flavonoids of O. glabra by gavage at the dosages of 100,200 and 300 mg/(kg·d) . The effects on superoxide dismutase(SOD) ,glutathione peroxidase (GSH-Px) and catalase (CAT) activities in serum and liver and brain tissues were observed,as well as the contents of malonaldehyde (MDA) and lipofuscin (LPO) . Effects of the flavonoids of O. glabra on the survival time in hypoxia and the loaded swimming time of mice were also researched. [Results]The flavonoids of O. glabra could enhance the SOD,GSH-Px and CAT activities of mice serum and tissues in different degrees,and could extremely significantly reduce the MDA and LPO contents in mice. High-dosage flavonoids of O. glabra could extremely significantly prolong the survival time in hypoxia and the loaded swimming time of mice. [Conclusions] The flavonoids of O. glabra had relatively good anti-aging effects.

  • Preliminary Test of Chemical Components and Content Determination of Total Flavonoids in Different Parts of Gonocaryum lobbianum

    [Objective]To research the chemical components in the roots,stems,leaves and nut shells of Gonocaryum lobbianum (Merr.) Kurz. ,and to detect the contents of total flavonoids in different parts of G. lobbianum. [Methods]The chemical components in water extract, 95% ethanol extract and petroleum ether extract from different parts of G. lobbianum were researched by preliminary tests. With rutin as the reference substance,the contents of total flavonoids in different parts of G. lobbianum were detected by NaNO2-Al(NO3)3-NaOH colorimetric method. [Results]Different parts of G. lobbianum all had polysaccharides,amino acid,polypeptide,protein,alkaloid,sterides,triterpenes,volatile oil,phenols,tannin,coumarin,lactone and flavonoids. There might be organic acids in different parts of G. lobbianum. Besides,there were anthraquinone,cardiac glycoside and phenols in the roots and leaves; and anthraquinone,cardiac glycoside and phenols might be existed in the stems and nut shells. Saponin did not exist in any part of G. lobbianum. The contents of total flavonoids in the roots, stems,leaves and nut shells of G. lobbianum were 0. 028% ,0. 017% ,0. 033% and 0. 012% ,respectively. Among them,leaves had the maximum content of total flavonoids. [Conclusions]The types of chemical components were preliminarily obtained in the roots,stems,leaves and nut shells of G. lobbianum; and the contents of total flavonoids in different parts were detected,which provided references for the further research on G. lobbianum.

  • Literatures Revaluation of the Pharmacological Effects of Anemarrhena asphodeloides

    [Objective]To evaluate of the pharmacological effects of Anemarrhena asphodeloides,so as to determine its exact pharmacological effects. [Method] Published literatures of Anemarrhena asphodeloides were consulted and classified. According to modern test methods and observation indexes of pharmacology,to review related pharmacological action of Anemarrhena asphodeloides literature and,in order to make clear its pharmacological action. [Result] The following effects of Anemarrhena asphodeloides are confirmed: anti-inflammatory,anti-virus, anti-senescence,anti-dementia,anti-epilepsy and anti-hyperthyroidism,but others lack sufficient test data: antibacteria,anti-pyretic,anti-cerebral ischemia and antitumor. [Conclusion]The most important pharmacological functions of Anemarrhena asphodeloides are the treatment of diabetes mellitus,delaying senescence and antiviral effect.

  • Research Progress in the Chemical Constituents and Biological Activity of Leucosceptrum

    The paper presented a systematic study on the chemical constituents and biological activity of Leucosceptrum,from which more than sixty constituents were extracted out,including sesterterpenoids,flavonoids,phenylethanoid glycosides,acyl glycosides and phenols,etc. Among them some sesterterpenes showed a variety of activities,such as antifeedant,antifungal,inhibiting PEP and antiangiogenesis. Therefore, to carry out the studies of Leucosceptrum plants will be of great significance to discovering new medicinal components.
