  • Extraction Technology of Polysaccharides from Gracilaria lamaneiformis and Its Antioxidation Research

    [Objective]To select the extraction technology of polysaccharides from Gracilaria lamaneiformis and to investigate its antioxidation.[Methods]The extraction technology of polysaccharides from G.lamaneiformis and its antioxidation were researched by response surface method.The microwave-assisted extraction of polysaccharides from G.lamaneiformis was optimized based on response surface method.[Results]The optimal extraction technology of microwave-assisted extraction was as follows:495 W microwave power,1∶100 solid-liquid ratio and17 min extraction time.Under this condition,the extraction rate of polysaccharides from G.lamaneiformis was 33.11%.The regression equation of the extraction rate of polysaccharides from G.lamaneiformis(Y)was Y=22.00+6.78A+1.15B+3.63C+0.82AB+2.97AC+0.37BC-0.52A2-1.86B2-2.26C2.Results of the antioxidation of polysaccharides showed that polysaccharides had certain clearance effects on DPPH free radical.[Conclusion]This optimized model had extremely significant regression and relatively good fitting,which was of certain application value

  • Purification Technology of the Total Flavonoids from Polygonum knotweed L. by Macroporous Resins

    [Objective]To optimize the purification technology of the total flavonoids from Polygonum knotweed L.by macroporous resins.[Methods]With rutin as the reference substance,the content of total flavonoids in P.knotweed was detected by aluminium nitrate coloration method.Taking static adsorption capacity and static adsorption rate as the investigation indices,the adsorption and desorption effects of four macroporous resins on the total flavonoids of P.knotweed were compared.The optimal macroporous resin was selected;and the adsorption and desorption conditions were researched.[Results]HPD-100 macroporous resin was most suitable for the purification of total flavonoids from P.knotweed.The purification technology was as follows:2.5 g/L sample concentration,2 BV/h sample flow rate;2.5 BV sample volume,70%ethanol as the eluant,2 BV/h elution flow rate,and 5 BV eluant dosage.Under this technology,the concentration of total flavonoids in the purified products was 55.4%;and the extraction rate of total flavonoids reached 83.2%(n=3).[Conclusions]HPD-100 macroporous resin showed good purification effects on the total flavonoids of P.knotweed.The optimized technology was reasonable,feasible and stable.

  • Regulating Effects of Dragon's Blood on the Lung Injury Repair of Rats and Its Mechanism

    [Objective]To research the regulating effects of dragon's blood on the lung injury repair of rats and its mechanism.[Methods]Lung injury repair model of rats was established by dripping the bleomycin into trachea.Rats were given the dragon's blood by gavage during the process of lung injury repair.Lung tissues of rats were collected on the 14thand 28thdays.Tissue chip was adopted;HE and collagen staining and immunohistochemical analysis were carried out.The collagen,collagen I and inflammatory cells were quantitatively analyzed.The TGFβ1mRNA expression of lung tissue was researched by Real-Time Fluorescence Quantitative RT-PCR.[Results]Dragon's blood had significant inhibitory effects on the inflammatory degrees of lung tissues and the collagen overdeposition on the two days(P<0.05);IOD values reduced from(0.549 2±0.210 5)and(0.957 8±0.375 6)to(0.274 9±0.159 2)and(0.318 1±0.121 0),respectively.Dragon's blood significantly reduced the TGFβ1mRNA expression on the two days(P<0.05);and the IOD values decreased from(0.021 9±0.011 0)and(0.017 0±0.015 7)to(0.008 3±0.002 4)and(0.006 2±0.005 1),respectively.[Conclusions]Dragon's blood could restrict the inflammatory injury of lung tissues,promote its repairing,and regulate the excessive repairing of mesenchyme through inhibiting the overdeposition of collagen,especially collagen I.The changes of collagen and collagen Imight be related to the expression decrease of TGFβ1mRNA in lung tissues.

  • Changes of the Physiological and Berberine Content in Phellodendron amurense Seedlings under Drought Stress

    [Objective]To research on the changes of the physiological indexes and berberine content in Phellodendron amurense seedlings under drought stress.[Methods]With P.amurense seedlings as the research materials,PEG 8000 with three different concentrations was used to simulate the drought stress.With MDA content,proline content and SOD activity as the investigation indexes,changes within 9 hours of drought stress were investigated.And the berberine content in leaves was detected.[Results]Under the stresses of 5%,10%and 20%PEG,changes of MDA content varied.Under 5%PEG treatment,MDA content reduced,and was lower than that before stress during the whole treatment(P<0.05).Under 10%PEG treatment,MDA content increased a little and a significantly increase appeared at the 9thhour(P<0.05).Under 20%PEG treatment,MDA content showed a single peak curve.Plasma membrane permeability showed a fluctuated upward trend as the stress time prolonged,and high PEG concentration led to great damage degree to membrane.Under 20%PEG treatment,proline content firstly decreased and then increased.Under a higher concentration of PEG,proline content varied greatly.SOD activity firstly enhanced and then reduced.With the increase of PEG concentration,the peak of SOD activity came earlier.Berberine content in leaves firstly enhanced and then decreased gradually as the stress time prolonged.Under higher concentration of PEG,the berberine content reached its peak earlier;while under lower concentration of PEG,the berberine content peak came later,but the maximum value was higher.[Conclusions]Drought stress had harmful effects on the physiology of P.amurense seedlings,but could enhance the berberine content in leaves.

  • Extraction and Purification Technologies of Total Saponins from RADIX ROSAE LAEVIGATAE

    [Objective]To select the optimal extraction technology of total saponins from RADIX ROSAE LAEVIGATAE;and to purify the total saponins by macroporous adsorption resins.[Methods]With the content of oleanolic acid as the reference substance,the total saponins content in RADIX ROSAE LAEVIGATAE was detected by UV-Vis spectrophotometry.Single factor test was adopted.The effects of extraction time,solid-liquid ratio and ethanol concentration on the total saponins of RADIX ROSAE LAEVIGATAE were investigated.Orthogonal test was designed according to the test results of single factor test.Purification effects of different types of macroporous resins on the total saponins of RADIX ROSAE LAEVIGATAE were evaluated according to the properties of static and dynamic adsorption and desorption properties.[Results]The influencing degrees of factors affecting the extraction rate of total saponins were in the order of solid-liquid ratio>reflux time>ethanol concentration.The optimal extraction technology was 1 h reflux time,1∶10 solid-liquid ratio,70%ethanol concentration,and two extraction times with 1 h each time.NKA-9 macroporous resin had the optimal purification effects on the total saponins of RADIX ROSAE LAEVIGATAE,and the purity of total saponins enhanced from 36.8%to 62.9%,and the elution rate reached 87.8%.[Conclusions]This method was effective,simple and feasible.

  • Extraction Technology of Polysaccharides from the Leaves of Coix lacryma-jobi L. by Cellulase Method

    [Objective]To optimize the extraction technology of polysaccharides from the leaves of Coix lacryma-jobi L.by cellulase method.[Methods]Effects of pH value,cellulase dosage,extraction time and hydrolysis temperature on the extraction rate of polysaccharides were researched by single factor test and orthogonal test.[Results]The optimal extraction technology of polysaccharides in C.lacryma-jobi leaves was as follows:was 5 pH,50℃hydrolysis temperature,2.5 h extraction time,and 500U cellulase in 1 g powder of C.lacryma-jobi leaves.Under this extraction technology,the average extraction rate of polysaccharides was 14.45%and RSD was 0.18%.[Conclusions]Extracting the polysaccharides from C.lacryma-jobi leaves by cellulase method had the advantages of time saving and high efficiency.

  • Dynamic Changes in Several Physiological Indices of Viola yedoensis Makino under Abiotic Stress

    [Objective]To determine the dynamic changes in several physiological indices of Viola yedoensis Makino under salinity,cadmium(Cd)stress and drought stresses,so as to deducing its physiological mechanism and laying foundation for further investigating the molecular mechanism and functional genes in response to abiotic stresses.[Method]Employing both hydroponic culture and potted cultivation methods,different salt concentration,Cd concentration and soil water content were set up on the plants with similar growth vigor for measuring MDA content,POD and SOD activities at various growth periods.[Results]Stressed by 0.4%Na2SO4,180μmol/L Cd and 12%of soil moisture,the physiological indices investigated showed regular change trends,indicating the graded response process of V.yedoensis to environmental factors.The activities of protective enzymes were the highest at 54 h,30 h and 3 d after salinity,Cd and drought stress respectively,when the expression of regulatory genes may peak in response to stress.[Conclusion]Whether MDA can be used as a physiological index for evaluating drought and salinity resistance of V.yedoensis still needs further experimental evidence,while the synergistic effect of protective enzymes POD and SOD can prevent cell membrane of V.yedoensis from damage caused by Cd and drought stresses.

  • Determination of Araloside X in Aralia elata( Miq.) Seem. by HPLC

    [Objective]To detect the Araloside X content in Aralia elata(Miq.)Seem.by HPLC.[Methods]Chromatographic column was Kromasil C18column(250 mm×4.6 mm,5μm);mobile phase was acetonitrile-0.05%phosphoric acid solution(35∶65,V/V);detection wavelength was 203 nm;column temperature was 30℃;flow rate was 0.8 mL/min;and injection volume was 10μL.[Results]Araloside X showed good linear relationship with peak area(r=0.999 9)within the range of 0.60-4.80μg/mL.The average recovery rate was 98.14%and RSD was 1.13%.[Conclusions]This method was sensitive,accurate and repeatable,and could be used for the quality control of A.elata.

  • Extraction and Identification of Ursolic Acid in HERBA VERBENAE

    [Objective]To research the preparation technology of ursolic acid in HERBA VERBENAE,and to establish its content detection method.[Methods]Ursolic acid was extracted and isolated from HERBA VERBENAE by ethanol extraction and agglutination separation.And content detection was established by the method of HPLC.[Results]After the extraction and isolation,the sample was further identified by spectroscopy,and was verified to be ursolic acid.The purity of ursolic acid reached 99.8%.When detecting its content by HPLC,ursolic acid showed good linear relationship within the range of 4.6-23.0μg/mL(r=0.999 9);and the average recovery rate was 99.82%(RSD was0.75%).[Conclusions]This preparation technology was advanced,rational,practical,feasible,and could be applied in the industrialized production.Detecting the content of ursolic acid by HPLC method was accurate and reliable.

  • Chemical Components of the Volatile Oils in Toddalia asiatica( L.)Lam in Different Production Areas

    [Objective]To discuss the differences in the chemical components of the volatile oils in Toddalia asiatica(L.)Lam in different production areas of Guangxi Zhuang Autonomous Region.[Methods]Volatile oils were extracted from T.asiatica by steam distillation method.And qualitative and quantitative analyses of volatile oils were analyzed by GC/MS.[Results]In the chemical components in the volatile oils of T.asiatica in four production areas of Guangxi,a total of 52 peaks were isolated from the T.asiatica in Nanning.Among them,34chemical components were identified.A total of 48 peaks were isolated from the T.asiatica in Liuzhou;and 29 chemical components were identified.A total of 43 peaks were isolated from the T.asiatica in Guilin;and 28 chemical components were identified.A total of 58 peaks were isolated from the T.asiatica in Bobai;and 40 chemical components were identified.Among them,there were 18 common peaks.The common chemical components in T.asiatica in Nanning,Liuzhou,Guilin and Bobai accounted for 81.88%,86.6%,73.09%and 81.29%,respectively.[Conclusions]The types of characteristic components showed no significant differences in the volatile oils of T.asiatica in four production areas of Guangxi,indicating that there were no differences in the germplasms of T.asiatica in four production areas.But the significant differences in the contents of chemical components showed that the soil and climate conditions in different production areas had relatively great effects on the plant growth of T.asiatica.

  • Ferulic Acid Content of Angelica pubescens in Three Gorges Reservoir Area

    [Objective]To study the quality standard of angelicae pubescentis radix,which is a genuine medicinal material of Three Gorges Reservoir Region,focusing on the establishment of the content determination method for ferulic acid in angelicae pubescentis radix.[Method]Using the ferulic acid content as the index,the ferulic acid content was determined by HPLC method,the optimum chromatographic conditions for the chromatographic column was TSK-GEL DDS-C18(4.6 mm×150 mm,5μm),mobile phase of anhydrous methanol-water-phosphoric acid(30∶70∶0.3,V/V/V),the flow rate of 1.0 mL/min,the detection wavelength of 320 nm,and the column temperature of40℃.[Results]The ferulic acid showed a good linear relationship in the range of 0.006-0.15μg/mL,and the regression equation was y=4 564 754x-(3 882.699,r=0.999 9),and the RSD of precision test was 0.67%;the RSD of peak area of stability test was 1.11%;the contents of 6 parallel ferulic acid samples were obtained by repeatability test,and the RSD was 1.20%;the contents of ferulic acid of a batch of concentrated Angelica were determined for 6 times in reproducibility test and the RSD was 0.93%;the recovery rate of 6 sampling obtained by recovery test was between 94.63%-99.51%,and the RSD was 1.67%,is less than 3%,indicating that the method is reliable.[Conclusion]the determination method for the contents of ferulic Angelica pubescens medicinal acid.The method is simple,rapid and accurate,reproducible,and effective control of Angelica pubescens medicinal materials quality.

  • Content Determination of Plumbagin in Plumbago zeylanica Flowers and Fruits

    [Objective]The content of plumbagin in Plumbago zeylanica L.flowers and fruits were determined,to provide the experimental basis for the clinical application and development of Plumbago zeylanica L.flowers and fruits.[Methods]The medicinal powders were immersed in cold methanol(50 mL)for 24 hours,and then extracted for 50min by ultrasound,to determine the content of plumbagin in Plumbago zeylanica L.flowers and fruits by HPLC.[Results]The content of plumbagin in Plumbago zeylanica L.flowers and fruits were respectively0.005 9%and 0.009 5%.[Conclusion]Plumbago zeylanica L.flowers and fruits contained plumbagin,which is the active ingredient,having the value of development and utilization.

  • HPLC Analysis for Fingerprinting of Bidens Genus Plants

    Objective]To establish an HPLC method for fingerprint of Bidens genus plants,for distinguishing five species of this genus.[Methods]Ten batches of Bidens genus medicinal materials were used for HPLC analysis.The chromatic parameters were as follows:Kromasil C18(4.6 mm×250 mm,5μm)as chromatic column,absolute methanol-0.1%phosphoric acid as mobile phase(gradient dilution),flowing speed at 1 mL/min,detection wavelength at 290 nm,column temperature at 30℃and injection amount of 10μL.[Results]The HPLC fingerprint for discriminating Bidens genus plants was established by using similarity software analysis.The medicinal materials tested were classified into five categories by primary ingredient analysis and phylogenetic analysis.[Conclusion]This method is simple,stable,repeatable and feasible,and it can well separate most of the chemical ingredients,which provides scientific and efficient method for discriminating Bidens genus medicinal materials.

  • Comparison of Different Drying Methods for the Rhizomes,Stems and Leaves of Ligusticum chuanxiong Hort.

    [Objective]To compare the effects of different drying methods on the effective components in rhizomes,stems and leaves of Ligusticum chuanxiong Hort.,which were baking after drying in the sunlight,natural drying in the shade and natural drying in the sunlight.[Methods]With butylphthalide content as the evaluation index,HPLC was adopted.Chromatographic conditions were as follows:chromatographic column was Diamonsil C18column(4.6 mm×250 mm,5μm);mobile phase was acetonitrile-0.4%acetic acid(55∶45,V/V);flow rate was 1.0 mL/min;detection wavelength was 228 nm;column temperature was 25℃;and injection volume was 10μL.[Results]Butylphthalide had good linear relationship(r=0.999 9)with peak area within the range of 0.515 2-5.152 0μg/mL.Limit of quantitation was 5.515 2 mg/L;average recovery rate was 100.10%,and RSD was 1.10%(n=9).As for the same drying method,butylphthalide content was in the order of rhizomes>stems>leaves.As for the different drying methods,baking after drying in the sunlight had the maximum butylphthalide contents in rhizomes,stems and leaves,followed with natural drying in the shade.And natural drying in the sunlight had the minimum butylphthalide contents.Butylphthalide content in L.chuanxiong by baking after drying in the sunlight had extremely significant differences with those by natural drying in the shade and sunlight(P<0.01).[Conclusions]Butylphthalide contents varied in the rhizomes,stems and leaves of L.chuanxiong.And baking after drying in the sunlight was more suitable for the whole plant drying of L.chuanxiong.

  • GC-MS Analysis of the Low-polarity Chemical Constituents from Parochetus communis Buch.

    [Objective]To study the low-polarity chemical constituents in the whole plant of Parochetus communis Buch.[Methods]The low-polarity chemical constituents were extracted from the whole plant of P.communis by organic solvent,and were isolated and identified by GC-MS method.The relative contents of the chemical constituents were determined by area normalization method.[Results]A total of 75peaks were detected and 61 compounds were separated and identified,which accounted for 98.78%of the total low-polarity chemical constituents.The main low-polarity chemical constituents were as follows:methyl palmitate(25.15%),pentadecane(5.93%),tetradecane(5.65%),methyl-9,12,15-octadecatrienoate(5.60%),clionasterol(3.84%),palmitic acid(3.83%),tridecane(3.74%),phytol(3.40%)and methyl linoleate(3.32%).[Conclusions]This research provided theoretical foundation for the further development and utilization of P.communis.

  • Advances in Pharmacological Activity of Febrifugine and Its Derivatives

    The herbal Dichroa febrifuga Lour is the dried roots of Saxifragaceae plant species Dichroa febrifuga Lour that are endowed with the efficacies of inducing vomiting,insecticide,anti-malaria and relieving heat.It has been broadly applied for treating malaria in folk.We reviewed the advances in pharmacological activity of effective component febrifugine in Dichroa febrifuga Lour and its derivatives,and found that Dichroa febrifuga Lour has been documented effective in antitumor,anti-coccidium,anti-scar,anti-malaria,and the treatment of diabetes mellitus and osteoporosis as well.On the basis of this,we proposed to develop safe and efficient derivatives continuously by structural modification and perform immune study and their antitumor activity.

  • Research Progress in the Pharmacological Functions of APS for Cardiovascular Diseases

    As an effective component in the traditional Chinese medicine of RADIX ASTRAGALI,Astragalus polysaccharides(APS)has the functions of enhancing immunity,reducing blood sugar,strengthening heart,anti-tumor,anti-virus,etc.Based on the relevant literatures in latest years,the paper introduced the pharmacological functions of APS in treating the cardiac diseases from four aspects,including the cardiovascular,cardiomyocytes,arterial pressure and heart rate.It has been proven that the APS could effectively improve the metabolites and chronic complications of diabetes patients,as well as regulate the blood sugar and protect the heart,lung and liver.However,given the little studies in this aspect,it deserves more and deeper discussions.

  • Advances in Chemical Components and Pharmacological Actions of Radix Fici Simplicissae

    Radix Fici simplicissae is the dried roots of Moraceae plant Ficus simplicissima that is endowed with the efficacies like spleen dampness,moistening lung for arresting cough and relaxing the muscles and joints.It has been one of folk medicines widely used in China for a long time.Here we review the research progresses in the chemical components and pharmacological action of Radix Fici simplicissae.Based on the reviews,we found that Radix Fici simplicissae is effective in treating the symptoms of swollen spleen,poor appetite,hepatitis,arthralgia due to wind-dampness and tuberculosis,and it has the functions of insecticidal,anti-oxidation and radioresistance,as well as the actions of protecting digestive system and enhancing immunity.Therefore,we apply modern S&T science and technology approaches to study Radix Fici simplicissae from quality assessment,effective components and action mechanism,aiming at providing reference for exploiting and utilizing the resources of Radix Fici simplicissae.

  • Clinical Evaluation on the Effect of Compound Wubeizi Hoop Ointment on Psoriasis Vulgaris

    [Objective]Discuss the clinical effect of compound Wubeizi hoop ointment on psoriasis vulgaris.[Method]104 cases definitely diagnosed with psoriasis vulgaris were grouped into two groups randomly,observation group(52 cases)and control group(52 cases).The observation group was treated with compound Wubeizi hoop ointment while the control group was treated with tretinoin(Vitamin A Acid).Then compare the clinical curative effects of the two groups.[Result]After 3 months of treating,the PASI score of the observation group was distinctly lower than that of the control group,and PASI scores of the two groups were significantly different(P<0.05).The total effective rate of the observation group reached 84.6%which was evidently higher than that of the control group,73.1%(P<0.05).[Conclusion]The clinical effect of compound Wubeizi hoop ointment on alcoholic fatty liver disease is good,so it is worthy of clinical application.
