  • Research on the Application and Current Situation of Anti-tumor by Traditional Chinese Medicine Volatile Oil Based on the Compilation of Chinese Medicine Standard Database

    The volatile oil is one of the most important active ingredients in traditional Chinese medicine( TCM). In recent years,research on the anti-tumor by TCM volatile oil is concerned greatly,such as screening of anti-tumor active site,excavation of anti-tumor substance basis and action mechanism,the development of anti-tumor preparation of TCM volatile oil,these are the foci of current research. According to the Compilation of Chinese Medicine Standard database,we selected 20 kinds of TCM volatile oil which has the anti-tumor effect,and applications of the 20 kinds of volatile oil in Chinese patent medicine,content and major component of volatile oil in Chinese herbal medicine,the effective extraction part and type of anti-tumor,granted patent and listed product were comprehensively analyzed. The research could provide a theoretical basis for in-depth study of the anti-tumor TCM volatile oil and systemic development of TCM volatile oil preparation.

Chemical Composition of Natural Medicine

  • A Study on Differences of Pigment and Nutrient Substance Contents among Impatiens L. Strains with Different Flower Colours

    [Objectives] The research aimed to study the differences of pigment and nutrient substance contents among Impatiens L. strains with different flower colours. [Methods] Nine Impatiens L. strains with different flower colours were used to examine the contents of pigments,non-structural carbohydrates and titratable acids of petal,leaf and stem by pot experiment,and their relativity was analysed. [Results]The redder the petal,the higher the anthocyanin content,with significant difference. The contents of flavonoid and total phenol in orange,light red and white petals were significantly lower than that of cardinal colour system of petal. The contents of carotenoid in pink,pale red and purplish red petals were significantly higher than that of other colour system of petal. But anthocyanin content was low in purple petal,meaning that main pigment in purple petal might not be anthocyanin. Moreover,a small amount of anthocyanin was detected in leaf and stem,and its content was higher in the strain with redder flower. Chlorophyll a content of leaf had no significant among test materials,whereas the contents of chlorophyll b and chlorophyll( a + b) of leaf from red petal strains were significantly higher than that of white and pale red strains. By correlation analysis,it was found that soluble sugar content of petal had significantly positive correlation with anthocyanin content; flavonoid content had significantly positive correlation with the starch and titratable acid contents of leaf,whereas showed very significant negative correlation with soluble sugar content of leaf. Relativity between total phenol and titratable acid was significant positive correlation in petal,and relativity between carotenoid content of petal and titratable acid content of leaf was very significant positive correlation,whereas showed very significant negative correlation with starch content of stem. [Conclusions] The research could provide theoretic basis for development and utilization of Impatiens L.

  • A Preliminary Study on the Chemical Constituents in Different Plant Parts of R. acetosa L.

    [Objectives] To conduct preliminary study on the chemical constituents in different plant parts of R. acetosa L. [Methods] The systematic pre-test method was used for physical and chemical identification of aqueous extract,ethanol extract and petroleum ether extract from different plant parts of R. acetosa L.,and the chemical constituents were preliminarily determined. [Results] The different plant parts of R.acetosa L. were positive in the identification test of carbohydrate,protein,phenol,organic acid,flavonoid and anthraquinone,indicating that these constituents may be contained; roots and leaves were negative in the identification test of saponins; the four plant parts of R. acetosa L.were negative in the identification test of steroid and triterpene in the ethanol extract,but positive in petroleum ether extract; roots and flowers were positive in identification test of volatile oil and fat. [Conclusions]By comparing the pre-test results about four different plant parts of R.acetosa L.,it was found that the total plant of R. acetosa L. might contain carbohydrate,glycoside,protein,phenol,organic acid,flavonoid and anthraquinone; the roots and flowers of R. acetosa L. might contain volatile oil and fat; stems and leaves might contain saponins.

  • Determination of Tracheloside and Trachelogenin in Trachelospermum jasminoides (Lindl.) Lem. from Different Production Areas by RPHPLC

    [Objective]To determine tracheloside and trachelogenin in Trachelospermum jasminoides( Lindl.) Lem. from different production areas by reverse phase high-performance liquid chromatography( RP-HPLC) method. [Methods] The experiment adopted ODS column( 4. 6mm × 250 mm,5 μm); the mobile phase was methanol-water solution; the volumetric flow rate was 1. 0 m L / min; the detection wavelength was 280 nm; the column temperature was 30℃. [Results]The highest content of tracheloside came from Chun'an of Zhejiang Province,followed by Tonglu of Zhejiang Province; the highest content of trachelogenin also came from Chun'an of Zhejiang Province,following by Lushan of Jiangxi Province. [Conclusions]The experiment results provide references for identification and application of Trachelospermum jasminoides( Lindl.) Lem.,and the established method is reliable,accurate and highly reproducible,and thus can be effectively applied in evaluating the quality of Trachelospermum jasminoides( Lindl.) Lem. from different production areas.

  • Study on the Changes of Chemical Components of Erjing Pills in the Preparing Process of Polygonatum sibiricum Delar

    [Objectives] The aim was to establish a UPLC method for simultaneous determination of two components in Erjing pills and compare the content changes of rutin and chlorogenic acid in the preparing process of Polygonatum sibiricum Delar. as well as optimize the compatibility of Erjing pills. [Methods] The chromatographic separation was carried out at 35 ° C on an ACQUITY UPLC HSS T3( 100 mm × 2. 1mm,1. 8 μm) column,the mobile phase was methanol( A) and 0. 2% phosphoric acid solution( B),the detective wavelength was 345 nm with the flow rate of 0. 4 m L / min. [Results]The contents of rutin and chlorogenic acid in Erjing pills after processing were 2- 3 times before processing. [Conclusions] The processed mixture of Polygonatum sibiricum Delar. and Lycium barbarum L. is the best,the method is simple,accurate and reproducible.

  • Determination of Chlorogenic Acid in Zhu Kuqin Granules by TLCHPLC Method

    The sensitive and simple methods of TLC and HPLC were developed for the identification and determination on chlorogenic acid of the novel compound Zhu Kuqin granule which was potential antidiarrheal compound granule of livestock and poultry. Chlorogenic acid was one of the active components in Zhu Kuqin granule existing in some plants or herbs. The chlorogenic acid identification of thin layer chromatography was achieved on H type of silica gel block using the upper solution of Butyl acetate-Formic acid-Water( 7:2. 5:2. 5,v / v / v) as developer,and the extraction of chlorogenic acid from Zhu Kuqin granule were sonicated with the methanol. The chromatography was performed on a Wonda Sil@ C18 column( 4. 6 mm × 250 mm,5 μm),and acetonitrile- 0. 4% phosphoric acid solution( 10:90,v / v,and adjusted to p H = 3. 0 with trithylamine) as the phase of CGA was used at the wavelength 324 nm. The results showed that the TLC of test sample had the same spots as that of reference chlorogenic acid. HPLC calibration curve of chlorogenic acid was linear in the range of 5 μg / ml- 200 μg / ml,and the average recovery rate was 98. 33%( RSD = 0. 587%,n = 6). The described methods were successfully applied to identification and determination of chlorogenic acid in Zhu Kuqin granule,which laid the foundation for further study of the granule of chlorogenic acid.

Pharmacological and Toxicological Analyses

  • Effect of Shenfu Injection on Serum C-reactive Protein,Calcitonin and TNF-α in Patients with Septic Shock

    [Objectives] To explore the influence of Shenfu injection on serum C-reactive protein,blood calcium and TNF-α in the patients with septic shock. [Methods]According to admission order,78 patients with septic shock were randomly divided into control and experimental groups. Control group was treated by isopropyl epinephrine,and experimental group was treated by Shenfu injection on the basis of the control group. [Results] The total effective rate was 95. 00% for the experimental group,obviously higher than that of control group( 78. 95%)with significant difference( P < 0. 05). After 3-d treatment,the experimental group's serum CRP and PCT levels were( 5. 83 ± 2. 51) mg / L and( 0. 97 ± 1. 12) μg / L,respectively,lower than the control group's PCT and CRP levels of( 10. 31 ± 2. 15) mg / L and( 4. 82 ± 1. 10)μg / L,with significant difference( P < 0. 05); the experimental group's serum TNF-α level was( 80. 83 ± 22. 51) ng / L,significantly lower than the control group's TNF-α level of( 184. 31 ± 28. 37) ng / L,with significant difference( P < 0. 05). [Conclusions] The Shenfu injection had a good therapeutic effect on patients with septic shock,and it could improve the level of CRP,PCT and TNF-α,so it could be promoted in clinic practice.

  • Protective Effect of Total Flavonoids Extracted from Polygonum knotweed L. on Mice with Liver Injury Induced by Antituberculosis Drugs

    [Objectives] To study the therapeutic effect and mechanism of total flavonoids extracted from Polygonum knotweed L. on mice with liver injury induced by antituberculosis drugs. [Methods]60 mice were randomly divided into six groups,namely normal control group,model group,glucurolactone group( 200 mg / kg),high-dose( 600 mg / kg),medium-dose( 300 mg / kg),and low-dose( 150 mg / kg) groups of total flavonoids extracted from P. knotweed. Except for the mice in the normal control group,the mice in the other groups were given intragastric administration of isoniazid and rifampicin( 100 mg / kg) to induce liver injury model,and were simultaneously treated with corresponding agents once a day for 30 d. On the 30 thday,the blood and liver tissue were sampled,and ALT,AST,MDA,SOD and GSH-PX content were detected by biochemical methods; pathological changes of liver tissue were observed after HE staining. [Results]Compared with the model group,ALT,AST and MDA content in the serum of the mice with liver injury increased,while SOD and GSH-PX content decreased in high-dose( 600 mg / kg) and medium-dose( 300 mg / kg) groups of total flavonoids extracted from P. knotweed( P < 0. 05 or P < 0. 01,and the pathological changes of liver tissue of the mice were alleviated at the same time. However,the low-dose( 150 mg / kg) group of total flavonoids extracted from P. knotweed had no significant effects on liver injury caused by antituberculosis drugs. [Conclusions]Total flavonoids extracted from P.knotweed has an obvious therapeutic effect on liver injury induced by antituberculosis drugs,and the mechanism may be associated with its antioxidative stress and inhibition of lipid peroxidation.

  • Study on the Antibacterial Activity of Several Medicinal Plants against Gray Mold

    [Objectives]To research the antibacterial effect of medicinal plant on gray mold. [Methods]The growth rate method was used to determine the antibacterial activity of ethanol extracts from eight kinds of medicinal plants against gray mold. [Results]The lilac,Amur Corktree bark and angelica extracts had a significant inhibitory effect on the mycelial growth of gray mold( P < 0. 05),the rate of inhibiting bacteria reached more than 60%,and EC_(50) was 0. 061,0. 065,0. 0743 mg / m L,respectively,significantly different from the antibacterial effect of several other kinds of medicinal plants; it was followed by Chinese goldthread,wild ginger and common cnidium fruit extracts,with EC_(50) of 0. 0976,0. 1516,0. 1821 mg / m L,respectively. [Conclusions]The medicinal plant extracts could be further applied and experimented in the production.

  • Dracorhodin Perchlorate Induced Apoptosis of Human Breast Cancer Cell MCF-7 through Mitochondrial Pathway

    [Objectives] To study anti breast cancer effect and action mechanism of Dracorhodin Perchlorate( DP). [Methods] MMT was applied to analyze inhibition rate of 60 μmol / L DP to cancer cells. Cell morphological analysis,cell nucleus morphological analysis,and DNA fragment analysis were carried out to determine occurrence of cell apoptosis. Rhodamine 123 staining was conducted to analyze mitochondrial membrane potential( MMP). Western blot was used to detect cell apoptosis correlation,especially expression of mitochondria related protein.[Results]60 μmol/L DP inhibited MCF-7 growth in time-dependent manner; DP inhibited MCF-7 growth through inducing MCF-7 apoptosis;when DP induced MCF-7 apoptosis,it activated mitochondrial pathway protein caspase-9,and PARP( poly ADP ribosepolymerase) expression was decreased after DP treatment. Further study indicated that major reason for DP inducing apoptosis is that it reduced MMP( in control group,93. 11% cells kept normal MMP,but in DP-treated group,only 37. 45% cells kept normal MMP). Mitochondrial Bcl-2 and Bcl-XL were decreased,and Bax and Bak increased,which led to drop of MMP. [Conclusions] DP induced apoptosis of human breast cancer cell MCF-7 through the mitochondrial pathway.

  • Effects of Gualou Xiebai Baijiu Decoction on Blood Lipid Content,Anti-oxidant Enzyme Activity and Inflammatory Factor Expression of Hyperlipidemia Mice

    [Objectives] To study the effects of Gualou Xiebai Baijiu Decoction( GXBD) on blood lipid content,anti-oxidant enzyme activity and inflammatory factor expression of hyperlipidemia mice. [Methods] 60 pieces of male Kunming variety mice were used. 10 pieces were randomly taken as normal standard group( NS),and the rest 50 pieces were fed with fat feeds. The hyperlipidemia model was established. According to blood lipid levels,they were divided into model standard group( MS),GXBD 13. 12 g / kg group,6. 56 g / kg group,3. 28 g / kg group,and Simvastatin( XFTT) 0. 004 g / kg group,each group had 10 mice,continuous administration was carried out for 8 weeks. In 24 h after final administration,TC,TG,HDL-C,CRP and LDL-6 in serum were measured,and SOD,GSH-Px,and MDA level in serum and liver tissue were measured. [Results] The TG,TC,and LDL-C level of the serum in model group were significantly higher than the normal standard group,while HDL-C level was significantly lower than the normal standard group( P < 0. 05). Besides,TG,TC,LDL-C,MDA,CRP,IL-6 level of serum and MDA level of liver tissue were significantly lower than the model group( P < 0. 05),while HDL-C,SOD,and GSHPx of serum and liver tissue were higher than the model group( P < 0. 05). [Conclusions] GXBD has significant function of adjusting blood lipid of hyperlipidemia mice,and the action mechanism is possibly related with its functions of improving anti-oxidant ability and inhibiting inflammatory reaction.

  • Function of CoQ10 in Regulation of e NOS in High ox-LDL Induced HUVECs

    [Objectives] To study the function of coenzyme Q10( Co Q 10) in regulation of endothelial nitric oxide synthase( e NOS) in high oxidized low-density lipoprotein( ox-LDL) induced human umbilical vein endothelial cells( HUVECs). [Methods] HUVECs in exponential growth phase were inoculated to 96 orifice plate at the cell density of 1 × 10~4 cells / 100 μL. When cells were fused to 80%,Tween-80 group and Co Q 10 group were randomly divided. After acting 24 hours,cell survival rate was tested by CCK-8 method,to screen cytotoxicity of Tween-80 and Co Q 10. HUVECs were cultured and divided into the control group,model group,Co Q10 low concentration group,Co Q medium concentration group,and Co Q high concentration group. After drug action for 24 hours,except the control group,a1 l groups were given culture solution containing 35 μg / ml ox-LDL to reproduce proliferation model of HUVECs. Changes in expression of e NOS mRNA level were detected by RT-PCR( reverse transcription polymerase chain reaction),and changes in expression of e NOS protein were detected by Western blot.[Results]Tween-80 dilution 10000 times had no cytotoxicity,and the difference was not statistically significant( P > 0. 05); when Co Q 10 concentration was 60 μM,it had cytotoxicity,and the difference was statistically significant( P < 0. 05). Compared with the control group,the e NOS in model group declined in expression of both the mRNA and protein,and the difference was significantly significant( P < 0. 05); compared with the model group,the e NOS mRNA and protein rose in Co Q 10 low concentration,medium concentration and high concentration groups,and the difference was statistically significant( P < 0. 05). [Conclusions]Co Q 10 can mitigate oxidative injury through improving the expression of e NOS in HUVECs.

  • Clustering Analysis of Tissues and Organs of Tibetan Medicines Renqing Mangjue Compatible with Zuota

    [Objectives] By clustering analysis of tissue distribution data of brucine and strychnine in gastric ulcer model rat,the impact of Zuota on tissue distribution of basic components was studied. [Methods] Based on system clustering method of SPSS19. 0 statistical analysis software,using inter-group join method and squared Euclidean distance,brucine and strychnine contents of different tissues and organs in nonZuota group and Zuota group were taken as characteristic variables for clustering analysis,and phylogenetic tree was established. [Results]When clustering distance was 1,( i) taking brucine content as the index,there were three kinds of convergences in non-Zuota group. A1class: skin,liver,epididymis and jejunum; A2 class: brain and uterus; A3 class: testis and muscle. Brucine contents of the three classes showed as A1 < A2 < A3. There were two classes of convergences in Zuota group. B1 class: jejunum,epididymis,kidney,brain,skin and uterus; B2 class: muscle and( bottom) submandibular gland. Brucine contents of the two classes showed as B1 < B2.( ii) Taking strychnine content as the index,there were three classes of convergences in non-Zuota group. C1 class: muscle,testicle and oarium; C2 class: heart and lung; C3 class: uterus and liver. Strychnine contents of the three classes showed as C3 < C1 < C2. There were two kinds of convergences in Zuota group. D1 class: kidney and heart; D2 class: brain tissue and uterus. Strychnine contents of the two classes showed as D1 > D2. [Conclusions]When clustering distance was 1,low-content tissues and organs firstly clustered,and its toxicological effect( or pharmacodynamic action) was insignificant,and this kind of tissues and organs were relatively safe. A1 class and A2 class in Zuota group were merged into B1 class,in which liver was replaced by kidney. It illustrated that Zuota could decline the toxicity of kidney,and enlarged the safe action range of brucine. Kidney and heart in C2 class were clustered into D1 class,and average strychnine content in C2 class was higher than that of D1 class. It could be deduced that Zuota had the effect of protecting heart.

  • The Effects of Alcohol and Water Extracts of Allium wallichii Kunth on Transplanted Hepatoma,Lung Cancer and Breast Cancer of Mice

    [Objectives] To study the anti-tumor effects of alcohol and water extracts of A. wallichii Kunth. [Methods]In vivo experiment of transplanted tumors was employed to observe the effects of different solvent extracts of A. wallichii Kunth on transplanted S180 sarcoma and transplanted H22 hepatoma of KM mice,transplanted Lewis lung cancer of C57 BL mice and transplanted EMT-6 breast cancer of BALB / C mice. 20 g / kg of different solvent extracts of A. wallichii Kunth were injected in mice with different types of transplanted tumors,while the blank control group was treated with same-dosage normal saline. After 11 days,the mice were killed,and the inhibitory rate of tumor was calculated. [Results] Compared with blank control group,the tumor weight of medicated group decreased obviously( P < 0. 05 or P < 0. 01),and the group treated with the extracts of roots and rhizomes,and leaves of A. wallichii Kunth had more obvious decrease effect in the same dosage( P < 0. 05 or P < 0. 01). [Conclusions] Different solvent extracts of A. wallichii Kunth showed anti-tumor effect,especially the extracts of roots and rhizomes,leaves of A. wallichii Kunth showed the obvious effect.

Processing of Traditional Chinese Medicine and Quality Control

  • Separation and Purification of Alkaloids in L. franchetii Beauv. by High Speed Counter-current Chromatography

    [Objectives] To develop a method for separation and purification of alkaloids from 95% ethanol extract of L. franchetii Beauv. by high speed counter-current chromatography. [Methods]Ultra performance liquid chromatography method was used to analyze and optimize solvent system and chloroform: methanol: water( 8:1:9) was selected as the solvent system of chloroform. The upper phase was used as the stationary phase,while the lower phase was used as the mobile phase with flow rate of 2 m L· min- 1,revolution speed of 900 r ·min- 1,separation temperature of 35℃,detection wavelength of 254 nm and injection volume of 20 m L. [Results] Aripiprazole( 39. 6 mg) was obtained from sample( 1. 0 g) extract and the purity was above 98. 51%. This alkaloid was found from plants for the first time. [Conclusions] This method was feasible and rapid,and could be used for separation and purification of alkaloids in L. franchetii Beauv.
