  • Review of Factors Influencing Active Components of Lonicera japonica Thunb.

    Lonicera japonica Thunb. is a typical medicinal and edible plant in China. It has antibacterial, anti-oxidant, anti-endotoxin, and anti-inflammatory effects, etc. Its pharmacological action is closely related to its active components. Based on the results of previous studies, this paper summarized the effects of variety, origin, cultivation conditions, pruning, harvesting period and processing methods on the active components of L. japonica, in the hope of providing theoretical guidance for high-yield and high-quality cultivation of L. japonica.

  • Protection of Intellectual Property and Authenticity of Chinese Herbal Medicine Chrysanthemum morifolium

    This paper first introduced Chrysanthemum morifolium Ramat. and its medicinal value. Then, it studied agricultural intellectual property resources such as geographical indications, trademarks, patents, new plant varieties, biological genetic resources, and traditional knowledge related to C. morifolium. Finally, it summarized poverty-stricken areas ecologically suitable for planting C. morifolium, analyzed the GAP certification of C. morifolium, pollution-free agricultural product certification, green food certification, and organic food certification, and discussed the strategies for C. morifolium intellectual property protection and authenticity protection in the context of TCM development strategy and rural revitalization strategy.

Chemical Composition of Natural Medicine

  • Chemical Components of Achyranthes bidentata Leaves by Ultra High Performance Liquid Chromatography-Mass Spectrometry

    [Objectives] To study the chemical components and relative content of Achyranthes bidentata leaves and provide a scientific basis for further development and utilization of A. bidentata leaves. [Methods] The chemical components of A. bidentata leaves were rapidly analyzed using the ultra high performance liquid chromatography-time of flight-high resolution mass spectrometry(UHPLC-TOF-MS). [Results] Thirty eight chemical compounds were identified in samples of A. bidentata leaves collected from Wen County of Henan Province, in which seven chemical compounds had the relative content higher than 5%, linoleic acid reached 25.7% and inokosterone A reached 13.8%. [Conclusions] A. bidentata leaves contain many kinds of chemical compounds. This study is expected to provide a certain basis for further extraction of linoleic acid and inokosterone A.

  • Simultaneous Determination of Six Constituents in Cangbaiqutong Capsule by HPLC Wavelength Switching Technique

    [Objectives] To establish a high performance liquid chromatography(HPLC) wavelength switching method for the simultaneous determination of content of six constituents(phellodendrine chloride, gentiopicrin, paeoniflorin, tetrandrine, berberine hydrochloride and paeonol) in Cangbaiqutong capsules, and provide a scientific basis for the comprehensive evaluation of the quality of Cangbaiqutong capsule. [Methods] The chromatographic column of Waters XSELECT CSH-C_(18)(4.6 mm × 150 mm, 5 μm), the mobile phase of acetonitrile-0.1% phosphate solution, gradient elution(0-15 min,10%-18% A;15-30 min,18%-50% A; 30-35 min, 50%-10% A); the flow rate of 1.0 mL/min, wavelength switching of 284(0-7 min, phellodendrine), 274(7-10 min, gentiopicrin), 230(10-14 min, paeoniflorin) and 274 nm(14-35 min, tetrandrine, berberine hydrochloride, paeonol), the injection volume of 10 μL. [Results] There was a good linear relationship between the area of chromatographic peak and the injection volume of phellodendrine chloride, gentiopicrin, paeoniflorin, tetrandrine, berberine hydrochloride and paeonol in the range of 0.150-1.504, 0.768-7.680, 1.096-10.960, 0.220-2.200, 0.296-2.956, 0.0345-0.345 μg, respectively; the average recovery rates(n=6) were 98.3%, 99.2%, 98.8%, 98.8%, 99.1% and 98.2%, respectively; the RSD value was 1.32%, 1.46%, 1.08%, 1.31%, 1.26% and 1.21%, respectively. [Conclusions] The method can be used to determine many kinds of constituents at the same time, and the operation is simple, accurate and reproducible, and can be used for the quality control of compound Cangbaiqutong capsules.

Pharmacological and Toxicological Analyses

  • Anti-inflammatory Effect of Wumen Zhike Gancao Decoction on Rats with Lumbar Disc Herniation Associated with Lipid Metabolic Disorder

    [Objectives] To study the anti-inflammatory effect of Wumen Zhike Gancao Decoction(ZKGC) on rats with lumbar disc herniation(LDH) associated with lipid metabolic disorder. [Methods] A rat model of LDH was established by implantation of the autologous nucleus pulposus from the coccygeal vertebra of each rat tail, and histopathology, immunohistochemistry and biochemistry assays were employed to evaluate the treatment effects of ZKGC. In addition, the metabolic characteristics of LDH and ZKGC treatment were investigated with a liquid-chromatography with time-of-flight mass spectrometer(LC/Q-TOF-MS)-based metabolomics study. Nucleus pulpous tissues from rat models were collected and analyzed by metabolomics. [Results] By metabolism network analysis, lipid metabolism was up-regulated in LDH rat models and the treatment with ZKGC significantly reversed the abnormal up-regulated lipid metabolism. Meanwhile, the treatment of ZKGC also regulated the markers of neuron autophagy and inflammatory response in serum. [Conclusions] These results indicated that a complex mechanism, including abnormal lipid metabolism, associates with the progress of LDH, and multiple pathways might be involved in ZKGC's therapeutic effects on LDH.

  • Anti-inflammatory Effects of Volatile Oil of Ocimum basilicum L. in Bozhou and Its Mechanisms

    [Objectives] The research aimed to study the anti-inflammatory effects of volatile oil from Ocimum basilicum L. in Bozhou through the experimental animal model of inflammation. [Methods] Xylene-induced mouse ear swelling model and carrageenan-induced mouse paw swelling model were used to compare the anti-inflammatory effects of volatile oil from O. basilicum L. [Results] The extremely high, high, medium, and low doses of volatile oil from O. basilicum L. had a certain inhibitory effect on the ear swelling of mice induced by xylene(P<0.05), and the inhibitory effect of high-dose group on the ear swelling of mice was better. The swollen degree of the mice's toes in the different dose groups of volatile oil from O. basilicum L. was significantly lower(P<0.05), and the high-dose and middle-dose groups had better inhibition. [Conclusions] The volatile oil from O. basilicum L. in Bozhou had a significant inhibitory effect on xylene-induced mouse ear swelling and carrageenan-induced mouse paw swelling. It proved and clarified the anti-inflammatory effects of volatile oil from O. basilicum L. in Bozhou.

  • Effects of Danqu Capsules on Arterial Blood Pressure,Heart Rate and Myocardial Tension Time Index in Rats

    [Objectives] To explore the effects of Danqu capsules(the prescription of Lei Zhongyi, the master of traditional Chinese medicine for the syndrome of intermingling of phlegm and static blood of coronary heart disease) on arterial blood pressure, heart rate and myocardial tension time index in rats. [Methods] A total of 50 SD rats, weighing(300±20) g, were randomly and evenly divided into Danqu capsule observation groups(low, middle and high-dose groups) and control groups(saline group and captopril group). Under anesthesia, the rats in each group were administered with 0.6, 1.2 and 2.4 g/kg of Danqu capsule suspension, 1 mL/100 g of saline and 10 mg/kg of captopril, respectively through duodena, and their systolic blood pressure, diastolic blood pressure, mean arterial pressure, pulse pressure difference and heart rate were measured and recorded by RM6240 multi-channel physiological signal acquisition and processing system before administration, 10, 20, 30, 40, 50, 60, 90 and 120 min after the administration, respectively, and the myocardial tension time index was calculated. [Results] Compared with the saline group, the systolic, diastolic and mean pressure reduced 10, 20, 30, 40, 50, 60, 90 and 120 min after the administration, and the pulse pressure difference did not change significantly in the Danqu capsule groups(low, middle and high-dose groups), and the heart rate slowed down and the myocardial tension time index reduced in the high-dose Danqu capsule group. Compared with the captopril group, the Danqu capsule groups showed similar blood pressure-reducing effect. [Conclusions]Danqu capsules have obvious effect of lowering blood pressure and myocardial tension time index. High-dose Danqu capsules has the effect of slowing down the heart rate. The blood pressure-lowering effect of Danqu capsules shows a dose-dependent manner. Clinically, they can be used in patients with coronary heart diseases and hypertension, suggesting that they should be used with caution in patients with hypotension.

  • Protective Effect of GRK2 and Effect of Sanguis Draconis Flavones on Focal Cerebral Ischemia-Reperfusion Injury in Rats

    [Objectives] To explore the protective effect of Sanguis Draconis flavones(SDF) on rat focal cerebral ischemia-reperfusion injury(CIRI) models established by middle cerebral artery occlusion(MCAO). [Methods] A total of 60 healthy adult male Sprague-Dawley rats were selected. They were evenly and randomly divided into sham group, model group, edaravone group(12 mg/kg) and SDF group(360 mg/kg), and administered intragastrically and intraperitoneally. The middle cerebral artery of each rat was blocked by suture-occluded method to establish a CIRI model. After ischemia for 2 h and reperfusion for 48 h, the pathological injury on the ischemic side was observed by HE staining; the neuron and myelin sheath structure was observed by transmission electron microscopy; the expression of G protein-coupled receptor kinase 2(GRK2) was preserved by immunohistochemistry; and the transfer of GRK2 was detected by western-blot. [Results] After 48 h of CIRI, the nuclei of the penumbral cortical neurons shrank, the chromatin was unevenly distributed, the nuclear membrane was dissolved and the mitochondria in the cytoplasm were swollen and vacuolated. The myelin layer was disordered. With this change, the distribution of GRK2 subcellular cells in the penumbra of the injured lateral cortex transferred from the cytoplasm to the membrane. SDF can effectively restore neuronal and myelin sheath structural damage and reduce the functional(membrane coupling) expression of GRK2. [Conclusions] GRK2 may be an effective target for SDF to protect the impaired blood-brain barrier(BBB) in CIRI.

  • Inhibitory Effects of Different Extractions of Salvia deserta Schang on Human Aldose Reductase

    [Objectives] To explore whether there are natural substances that inhibit human aldose reductase in different extracts of Salvia deserta Schang. [Methods] The inhibitory activities of human aldose reductase from 28 extracts were screened. [Results] The methanol extract of roots of Salvia deserta Schang had the strongest inhibitory effect on human aldose reductase. [Conclusions] It was confirmed that Salvia deserta Schang has inhibitory effect on human aldose reductase.

Processing of Traditional Chinese Medicineand Quality Control

  • Determination of Moisture, Ash, Extract Content and TLC identification of Tibetan Medicinal Material Dracocephalum tanguticum Maxim.

    [Objectives] The moisture, ash and extract content of Dracocephalum tanguticum Maxim. were measured. [Methods] The moisture, total ash, acid-insoluble ash and extract content of D. tanguticum Maxim collected from 16 producing areas were detected by using the methods in Chinese Pharmacopoeia(2015 edition, volume I), and D. tanguticum Maxim. was identified by thin layer chromatography in Chinese Pharmacopoeia(2015 edition, volume I). [Results] The moisture content of D. tanguticum Maxim collected from 16 producing areas ranged from 8.68% to 10.36%, averaging 9.28%. The content of total ash was between 7.21% and 11.60%, averaging 9.89%. The content of acid-insoluble ash was 0.21%-2.71%, averaging 1.51%. The extract content(water-soluble) was 26.67%-42.91%, averaging 32.65%. According to the results of TLC identification, the 16 samples and reference substance had the same characteristic spots at the corresponding positions. [Conclusions] It was recommended that the moisture, total ash and acid-insolue ash content of D. tanguticum Maxim were not be more than 11%, 12%, and 3% respectively, while the extract content was not less than 26%. It provides reference for filling the gaps in the quality standards of D. tanguticum Maxim.

  • Pressurized Synchronous Extraction of Volatile Oils, Total Flavonoids and Tannic Acid from Artemisia argyi

    [Objectives] This study aimed to optimize the conditions for pressurized synchronous extraction of volatile oils, total flavonoids and tannic acid from Artemisia argyi. [Methods] Single factor experiments and orthogonal experiments were conducted. [Results] Pressurization had a significant effect on the extraction rate of chemical constituents of A. argyi. The optimal conditions for pressurized synchronous extraction of volatile oils, total flavonoids and tannic acid from A. argyi were as follows: solvent concentration of 70%, solid to liquid ratio of 1∶30(g/mL), extraction pressure of 0.9 MPa, extraction time of 40 min and extraction temperature of 90 ℃. Under the optimal extraction conditions, the yields of volatile oils, total flavonoids and tannic acid were 0.852%, 4.66% and 6.79%, respectively. [Conclusions] Compared with other extraction methods, the pressurized assistant solvent extraction method can achieve the synchronous extraction of volatile oils, total flavonoids and tannic acid from A. argyi. The process is stable, short in extraction time, high in solvent utilization rate and ideal in extraction effect of the three components from A. argyi.

  • Determination of Gallic Acid in Rotala rotundifolia and Shuijia Zhixue Capsule by HPLC

    [Objectives] To establish a method for the determination of gallic acid in Shuijia Zhixue capsule and Rotala rotundifolia. [Methods] High performance liquid chromatography(HPLC) and Phenomenex C_(18) column were used, the mobile phase was 0.1% phosphoric acid∶methanol=85∶15, the flow rate was 0.5 mL/min, and the detection wavelength was 270 nm. [Results] In the range of(2-25) μg/mL, the concentration of gallic acid had a good linear relationship with the peak area, r=0.990 8; the average recovery was 96.88%. [Conclusions] The method was simple, sensitive and reproducible. It provided a basis for the analysis of gallic acid in Rotala rotundifolia and Shuijia Zhixue capsule and the establishment of quality detection methods in the future.

  • Phenotypic Diversity of Different Populations of Paeonia ludlowii in Nyingchi of Tibet

    [Objectives] To study the differences in fruit traits of different populations of Paeonia ludlowii,and to provide basic data for the identification,protection and utilization of germplasm resources of P. ludlowii.[Methods] Nine fruit traits of three different populations of P. ludlowii in Tibet were tested and analyzed.[Results]The mean values of 9 fruit traits of P. ludlowiiwere different in different populations;the coefficients of variation of different traits of different populations were also different; there were morphological differences of different sizes among individuals in each population; there were significant differences in 8 traits (pod length,pod width,pod thickness,pod weight,seeds per pod,seed length,seed width,and seed weight) among different populations. Among the 9 traits of the three P. ludlowii populations,the variability of the five traits was higher in the fruit pods,the variation degree of five traits including pod weight,pod thickness,seeds per pod,and seed weight was higher. It is of great significance for the screening and preservation of the germplasm resources of P. ludlowii. According to the principal component analysis (PCA),the seed weight,seed width,and seed length,i. e. the seed traits,were the main factors causing differences in fruit traits among populations. There was a certain correlation between different traits of P. ludlowii. Among the 36 Pearsonrelated combinations of 9 fruit traits,11 combinations had extremely significant correlations,and 8 combinations had significant correlations,indicating that there was a certain correlation between fruit traits in the growth and development. Through cluster analysis,three populations of P. ludlowii can be divided into two categories. The BV population was a category,which is characterized by long pods,long seeds,wide seeds,thick seeds and large seed weight. Therefore,the BV population is suitable material for breeding excellent varieties.[Conclusions]This study provides basic data for the identification,protection and utilization of germplasm resources of P. ludlowii.

  • Typical HPLC Chromatogram of Petroleum Ether in Abutilon indicum(L.) Sweet from Different Habitats

    [Objectives] To study the typical HPLC chromatogram of petroleum ether in Abutilon indicum(L.) Sweet from different habitats, in order to provide experimental basis for the quality control of A. indicum and the spectrum-efficacy relationship of petroleum ether. [Methods] The HPLC chromatogram of petroleum ether was obtained using Agilent SB-C_(18)(4.6 mm×250 mm, 5 μm) and acetine-0.1% phosphoric acid solution for gradient elution at temperature of 30 ℃, flow rate of 1.0 mL/min and detection wavelength of 205 nm. [Results] The typical HPLC chromatogram of petroleum ether in A. indicum was preliminarily established, with 12 characteristic peaks. It was verified by A. indicum from different habitats, characterized by reproducibility and stability. [Conclusions] Typical HPLC chromatogram provides a stable new method for the quality control of petroleum ether in A. indicum. This study provides experimental basis for further study on the medicinal parts of A. indicum.

  • Effects of Different Substrates on Seed Germination and Growth of Piptanthus nepalensis

    [Objectives] To find out the more suitable substrate for seed germination and seedling growth by comparing the effects of different substrates on seed germination. [Methods] Taking the seeds collected in Nyingchi Lulang area of Tibet as the research object, the germination effects(germination percentage, germination potential, germination index) and seedling growth status(primary root number, secondary root number, main root length, root-shoot ratio, seedling height) were measured and compared. [Results] The germination effect of Piptanthus nepalensis seeds was different in different substrates, and the germination rate, germination potential and germination index of P. nepalensis seeds in vermiculite were better than those in humus soil and perlite; the growth status of seedlings in different substrates was different, the number of primary and secondary roots in vermiculite substrate was significantly larger than that in the other two substrates, and the seedling growth in vermiculite was also better than that in the other two substrates; in comparing the main root length and root-shoot ratio, it was found that the main root length in perlite was longer than that in the other two substrates, and the root-shoot ratio was also higher than that in the other two substrates. [Conclusions] The seed germination effect and seedling growth status of P. nepalensis in different substrates were different, and there were significant differences. Vermiculite substrate had better effect on seed germination and growth, and vermiculite substrate could better promote seed germination and seedling growth. Vermiculite substrate was a good choice for seedling cultivation of P. nepalensis.

  • Study on Polyphenol Oxidase Activity and Total Phenol Content of Docynia indica

    [Objectives] To study the polyphenol oxidase (PPO) activity and total phenol content of Docynia indica.[Methods]The tender branches and petioles of the medicinal and edible plant D. indica were used as experimental materials,and the annual changes of total phenol content and polyphenol oxidase (PPO) activity were analyzed.[Results] pH7. 0 was the optimum value of polyphenol oxidase activity in D. indica plants; the best substrate was catechol with the concentration of 0. 15 mol/L; the optimum temperature was 25℃.[Conclusions]The total phenol content of D. indica plants was the lowest in April,May and September of each year. The polyphenol oxidase activity of D. indica plants at flowering and fruiting stage was significantly higher than that at pre-flowering stage.

Clinical Application

  • Study on the Cutting Propagation Technique of Himalayan Leycesteria formosa Wall. Twigs

    [Objectives] The study aimed to protect the good germplasm resources of Himalayan Leycesteria formosa Wall. and produce a large number of L. formosa plants used in Tibetan garden landscape. [Methods] In the intelligent solar greenhouse, the effects of different concentrations of ABT2 rooting powder on the cutting propagation of L. formosa twigs were studied, and the effects of different matrices on the growth of the cutting seedlings were analyzed. [Results] The effect of different concentrations of ABT2 rooting powder on the rooting rate of L. formosa is as follows: 200 mg/L>300 mg/L>100 mg/L, and the rooting rate is as follows: 61%>59%>57%. The rooting rate of L. formosa in the control group(CK) was 54%. The average number of roots and the average length of main root of L. formosa in the three treatments with ABT2 rooting powder were significantly different, while the average number of primary roots, average number of new branches, and average number of new leaves of L. formosa treated with 100 mg/L ABT2 rooting powder had no significant difference. There were significant differences between 200 and 300 mg/L ABT2 root powder treatments in the three growth indicators of L. formosa, and the promotion effects of 200 mg/L ABT2 rooting powder on the five indicators of L. formosa cuttings were better. The effects of different transplanting matrices on the indoor survival rate of L. formosa cutting seedlings were very obvious, and the survival rate of L. formosa cutting seedlings in matrix C was the highest, up to 96%. Different substrates had different effects on the growth of ground diameter of L. formosa cutting seedlings, and the promotion effect of matrix C was better. [Conclusions] In the rapid cutting propagation of good L. formosa, 200 mg/L ABT2 rooting powder can be selected. For indoor transplanting, matrix C(sandy soil+perlite+humus soil) should be used.

  • Clinical Study on Huangku Qingxin Mixture Combined with Metoprolol Tartrate Tablets for Treatment of Ventricular Premature Beat

    [Objectives] To observe the clinical effect and safety of Huangku Qingxin Mixture combined with Metoprolol Tartrate Tablets for patients with ventricular premature beat. [Methods] 126 patients were divided randomly into the control group(62 cases) and the treatment group(64 cases). The control group was treated with routine western medicine, and the treatment group was treated with routine western medicine combined with Huangku Qingxin Mixture, both for one month. Then, observed the changes of Myerburg score, SAS score, symptom score and dynamic electrocardiogram in the two groups before and after treatment. [Results] The total effective rate of clinical symptoms was 82.81% in the treatment group and 64.52% in the control group. Through comparison of two groups, the difference was statistically significant(P<0.05). The total effective rate of dynamic electrocardiogram was 84.38% in the treatment group and 58.06% in the control group. Through comparison of two groups, the difference was statistically significant(P<0.05). Before treatment, there was no significant difference being found in the comparison of the scores of Myerburg and SAS in the two groups(P>0.05). After treatment, Myerburg score and SAS score in the two groups were lower than those before treatment(P<0.05); Myerburg score and SAS score in the treatment group were both better than those in the control group(P<0.05). Before treatment, there was no significant difference in the comparison of the QT interval dispersion(QTd) and the corrected QT interval dispersion(QTcd) by Bazette formula(P>0.05). After treatment, the results of QTd and QTcd in the two groups were lower than those before treatment(P<0.05); the results of QTd and QTcd in the treatment group were better than those in the control group(P<0.05). [Conclusions] The application of Huangku Qingxin Mixture combined with Metoprolol Tartrate Tablets can effectively reduce Myerburg scores and SAS scores in patients with ventricular premature beat, shorten QTd and QTcd, and improve the life quality of patients. It has no adverse clinical reaction.

  • Effects of Onion Extract on Male Sexual Dysfunction Induced by SSRI Antidepressants

    [Objectives] To investigate the effect of onion extract on male sexual dysfunction caused by SSRI antidepressants. [Methods] 40 male patients with depression and anxiety who took SSRI antidepressants for a long time were randomly divided into observation group and control group. The observation group was treated with onion extract and SSRI drugs, while the control group was treated with SSRL drugs only. After 4 weeks of treatment, the scores of the two groups were evaluated by the International Index of Erectile Function(IIEF) questionnaire and the serum testosterone levels were measured in the two groups. [Results] Onion extract could improve the sexual dysfunction caused by SSRI drugs and significantly increase the level of serum testosterone. [Conclusions] Onion extract could significantly enhance the sexual function of men, and could alleviate the sexual dysfunction caused by SSRI drugs by increasing the level of serum testosterone.
